摘    要:目的探讨金黄色葡萄球菌(简称金葡菌)诱导人巨噬细胞系U937细胞凋亡时Fas/FasL蛋白的表达及意义。方法当细胞∶细菌为1∶20时分别培养0,15,30,60和90min,采用Western blotting检测Fas/FasL蛋白的表达情况,应用SPSS10.0软件包对结果进行统计学分析。结果Fas/FasL蛋白的表达随着金葡菌感染时间的延长而增高。结论金葡菌能够上调Fas/FasL蛋白的表达,从而可以诱导U937细胞的凋亡。

Abstract:[ Objective] The relationship between the induced apoptosis and expression of heat stress protein 90kd (HSP90) in human monocytic U937 cells by Staphylococcus aureus (S. aureus) was investigated. [ Methods ] U937 were used in all experiments, and they were infected with S. aureus when U937: S. aureus was 0, 1:5, 1:10, 1: 20, 1:50 and 1:100 respectively for 30 rain or for different lengths of time (0 min, 15 min, 30 min, 60 rain and 90 min respectively) when U937:S. aureus was 1:20. The apoptosis rates were examined by Annexin V FTTC/PI staining. Expression of HSP90 was detected by western blot. [Results] S. aureus could induce apoptosis of U937 in a dose- and time-dependent manner. The expression of HSP90 was upregulated in a time of infecting and dose of S. aureus dependent manner. [ Conclusions ] It is believed that U937 apoptosis induced by S. aureus may be related to the increase of HSP90.
