金黄色葡萄球菌对U937细胞HSP90蛋白表达的影响

摘    要:目的探讨金黄色葡萄球菌(简称金葡菌)诱导人巨噬细胞系U937细胞凋亡以及热休克蛋白90(HSP90)在这一过程中的作用。方法当U937:金葡菌分别为0,1∶5,1∶10,1∶20,1∶50和1∶100时,培养30min;当U937:金葡菌为1∶20时,分别培养0,15,30,60和90min。采用Annexin V FITC/PI双染流式细胞仪检测U937细胞凋亡率;采用Western blot方法检测HSP90蛋白的表达。结果U937细胞凋亡率随着金葡菌浓度的增加及感染时间的延长而逐渐增高;HSP90的表达随着金葡菌浓度的增加及感染时间的延长而逐渐升高。结论金葡菌诱导的U937的凋亡可能与HSP90蛋白表达的改变有关,并且具有感染时间及细菌浓度依赖性。

Abstract:【Objective】The relationship between the induced apoptosis and expression of heat stress protein 90kd (HSP90) in human monocytic U937 cells by Staphylococcus aureus (S. aureus) was investigated.【Methods】 U937 were used in all experiments, and they were infected with S. aureus when U937: S. aureus was 0, 1∶5, 1∶10, 1∶ 20, 1∶50 and 1∶100 respectively for 30 min or for different lengths of time (0, 15, 30, 60 and 90 min respectively) when U937:S. aureus was 1∶20. The apoptosis rates were examined by Annexin V FITC/PI staining. Expression of HSP90 was detected by western blot. 【Results】S. aureus could induce apoptosis of U937 in a dose-and time-dependent manner. The expression of HSP90 was upregulated in a time of infecting and dose of S. aureus dependent manner. 【Conclusions】It is believed that U937 apoptosis induced by S. aureus may be related to the increase of HSP90.
