摘    要:目的:研究牵拉培养对表皮细胞生长的影响以及内源性EGF的分泌情况。方法:用酶消化法对表皮细胞进行原代培养,将实验分为二组:实验组和对照组,实验组采用细胞负压牵引装置培养,对照组细胞不受牵拉,连续观察72小时,分别于12、24、36、48、60、72小时,收集细胞培养液,同时于72小时观察表皮细胞在牵拉条件下和非牵拉条件下的生长情况:用ELISA法测定表皮细胞在牵拉条件下和非牵拉条件下分泌EGF的情况。结果:实验组表皮细胞经72小时牵拉后生长良好,而对照组的细胞死亡,实验组牵拉36小时EGF浓度为16.03μg/mL,其他时间点未测到,而非牵拉组各时间点均未测出EGF。结论:牵拉不但可促进表皮细胞的增殖和生长,而且可促进表皮细胞分泌EGF.

Abstract:Objective To explore the effect of strain on the growth and secretion of EGF of human epidermal cells in vitro. Methods the study was performed with a vitro model of expansion in which epidermal cells were cultured on a flexible-bottom membrane that could be mechanically deformed by vacuum.Epidermal cells were respectively seeded equally on flexible membranes coated with type I collagen.The effect of cyclic strain on human epidermal cells in vitro was investigated.Cells were incubated for 72hr which were cultured in serum-free medium.The medium were collected respectively after 12-72 h of stretch.EGF concentrations were determined using an enzyme-linked immunosorbent assay. Results After 72h of stretch epidermal grew well.Conversely epidermal cells in the control group had died after 72h.EGF after 36h increased significantly,reaching 16.03pg/ml after 36h,and could not determined apart from 36h after stretch.Conversely,EGF could not determined during 12-72h in control group. Conclusion These findings suggest that cyclic strain promotes proliferation and secretion of EGF of cultivated epidermal cells.
