摘    要:以曲安奈德(TA)作为模型药物,壳聚糖和明胶作为基体,制备治疗眼穿通伤用的复合药膜(GICS-TA)。溶剂挥发法制膜,采用红外光谱,X-射线衍射、扫描电镜分析药膜的结构与组成,并植入青紫蓝兔眼脉络膜上腔观察其对眼穿通伤治疗效果。TA与载体有良好的相容性,活性完好。药膜与眼组织生物相容性好,药物可持续释放,通过脉络膜到达视网膜和玻璃体内,抑制眼穿通伤术后创伤性炎症和玻璃体增殖反应,治疗效果优于传统给药方式,可望成为治疗眼穿通伤的新方法。

Abstract:Using triamcinolone acetonide(TA) as a model drug and gelatin-chitosan as carriers,the drug membranes(GICS-TA) for ocular perforating injury were prepared.The drug films were prepared by solvent volatilization.Then structures and composition were analyzed by Fourier transform infrared spectroscopy(FT-IR),X-ray diffraction(X-RD) and scanning electron microscopy(SEM).Their cure efficacy for ocular perforating injury was observed after implanted into choroid cavity of blue-purple rabbits'eyes.There was good compatibility between drug and carriers.The drug activity was kept well.The films showed good biocompatibility and kept sustained drug release.The drug was carried into retina and vitreous body through choroid coat,restraining wound inflammation and vitreous body proliferation reaction after operation on ocular perforating injury.And the cure efficacy showed better than that of traditional administration.The drug-loading membrane is expected to be a new effective method for ocular perforating injury of in clinic in the future.
