2型糖尿病脂肪细胞胰岛素受体活性的研究

摘    要:目的:通过脂肪细胞胰岛素受体(ICIR)活性测定,探讨2型糖尿病(DM)部分发病机理。方法:Rodbell法获取游离脂肪细胞,放射受体分析法分别测定2型DM及正常对照组的ICIR。结果:两2型DM组高亲的力ICIR及低亲和力ICIR均明显低于正常对照组,其中低亲和力ICIR减少更甚(P<0.01)。2型DM伴高胰岛素血症组ICIR数量较不伴高INS血症组减少更甚(P<0.05)。两糖尿病组INS与受体亲和力减低。结论:2型DMICIR存在数量减少及质的改变,这种改变系原发性缺陷。INS对其受体有下降调节作用,这对2型DMICIR的改变起非主要的作用。

Abstract:Objective To investigate the defect of adipocytes insulin receptor and mechanism of type 2 diabetes mellitus(DM).Methods:We picked up isolated adipocytes with Rodbdll's method.We took the method of radioreceptor assay to determine insulin-binding in type 2 DM group and control guoup.Results:The numbers of low affinity receptor and high affinity receptor in both type2 DM grorps decreased significantly,more than that of normal control group.The reduced receptors in both type 2 DMgrorps were the receptors with low affinity and high capacity( P <0.01).The numbers of adipocyte insulin receptors in type 2 DM company heperinsulinemia decreased significantly more than that of type 2 DM no heperinsulinemia( P <0.05).Insulin receptor affinity was lowered in both type 2 DM group.Conclusion:There were the change of affinity and the decrease of insulin receptors in type 2 DM.The change is a primary defect.Down regulation of heperinsulinemia was not one of important causes of the defect of adipocytes insulin receptor in type 2 DM.
