骨巨细胞瘤中MMP-2的表达及其意义

摘    要:目的 探讨基质金属蛋白酶- 2(MMP -2)的表达与骨巨细胞瘤血管新生、肿瘤增殖、转移及复发的关系。方法 采用免疫组化法检测82例骨巨细胞瘤标本MMP- 2的表达,并分析其与肿瘤细胞增殖、血管形成及转移复发的关系。结果 骨巨细胞瘤中 MMP- 2 的阳性表达率为 60 .98%。MMP -2表达强度与微血管密度(MVD)值显著相关(P<0 .05),不同 MMP 2 表达组的复发率、转移率有显著性差异(P<0. 05),多因素Logistic回归分析显示MMP -2表达及手术方式对复发有显著影响(P<0 .05)。结论 MMP -2的表达与骨巨细胞瘤的血管生成、转移及复发有关,可作为判定骨巨细胞瘤转移及复发潜能的参考指标。

Abstract:Objective To evaluate the expression of matrix metalloproteinase-2(MMP-2) and its relationship to tumor angiogenesis,invasion and recurrence of giant cell tumors of bone(GCT).Methods The expressions of MMP-2,PCNA and CD34 in 82 cases of human GCT were examined using immunohistochemical staining,and intratumoral microvessel density was calculated.The expressions of MMP-2,PI value,and MVD were analyzed with their relations to the invasion and recurrence of the tumor.Results The expression of MMP-2 in GCT was 60.98%,and its expression was significantly associated with MVD(P<0.05).The rate of invasion and the rate of recurrence were significantly different between MMP-2 positive and negative groups(P<0.05).Multiple logistic regression analysis revealed that operation methods and MMP-2 were significantly related with recurrence(P<0.05).Conclusion The expression of MMP-2 is highly related to the invasion and recurrence of GCT,and can be used to predict the recurrence ability of GCT and guide the clinical therapy of GCT.
