ICAM-1在脊髓缺血再灌注损伤后表达变化及意义

摘    要:目的 探讨细胞间粘附分子 1在大鼠脊髓缺血再灌注损伤后表达变化及其意义。方法 采用Zivin法建立脊髓缺血再灌注损伤模型。以GAPDH为内参照物 ,应用半定量RT PCR方法、免疫组化及免疫荧光激光共聚焦显微镜定性、定量测定脊髓微血管内皮细胞表面ICAM 1mRNA表达变化。结果 正常脊髓微血管内皮细胞表面仅有微量的ICAM 1表达 ,单纯缺血不引起ICAM 1表达。再灌注后损伤区ICAM 1表达发生了改变 ;再灌注 4h ,ICAM 1mRNA表达量明显升高 ,再灌注 12h其在单位微血管面积上的荧光强度约比单纯缺血组增加了 1 2。提示脊髓损伤后微血管内皮细胞初始ICAM 1表达增加是由再灌注损伤激发 ,其后延迟递增表达增强 ,说明ICAM 1参与了脊髓再灌注损伤的炎症病理过程。结论 ICAM 1可做为脊髓I R后炎症反应程度的监测指标。

Abstract:Objective To investigate the significance and change of expression of mircrovascular ICAM-1 after spinal cord ischemia/reperfusion injury. Methods Set up spinal cord ischemia/reperfusion model. The expression of vascular endotheliocytes ICAM-1 mRNA in spinal cord injury was detected through the adoption of PCR, immunohistochemistry and immunofluorescence technique. Results The expression of adhesion molecules didn't increase in the normal group and the pure ischemia group.But the expression of adhesion molecules increased in ischemia area after reperfusion. The expression of ICAM-1 mRNA increased two hours, after reperfusion which was about twice as much as that of the comparative groups.It peak appeared 4 hours, after reperfusion and lasted for twelve hours twelve hours. After reperfusion its fluorescent intensity on microvascular area per unit was increased about 50% more than that of the ischemia group. Conclusion The ICAM-1 would be monitoring marker of Inflammatory reaction after spinal cord ischemia/reperfusion injury.
