肝硬化大鼠食管一氧化氮合酶分布定位研究

摘    要:目的探讨一氧化氮(NO)在肝硬化食管静脉曲张及血流动力学改变中的作用。方法应用还原型辅酶Ⅱ-黄递酶(NADPH-d)组化染色观察CCI4所致肝硬化大鼠食管下段一氧化氮合酶(NOS)分布;应用荧光法检测大鼠食管组织匀浆及血清中NO含量;应用57Co标记微球测定大鼠血流动力学指标。结果肝硬化鼠食管粘膜及粘膜下血管内皮NOS呈强阳性反应,而正常鼠则呈弱阳性或阴性反应;肝硬化大鼠食管及血清中NO含量显著升高。且全部出现门脉高压和高动力循环状态。结论NO在肝硬化大鼠食管静脉曲张及血流动力学改变中可能起重要作用。

Study on the localization of nitric oxide synthase in esophagus of cirrhotic rats
Abstract:Objective To investigate the effects of nitric oxide(NO) on esophageal varices and hemodynamic changes in cirrhotic rats. Metbods The nitric oxide synthase(NOS) distribution in lower esophagus of rats with carbon tetra- chloride-induced cirrhosis was studied by using NADPH-diaphorase (NADPH-d) histochemical method. Concentration of NO in esophageal tissue homogenate and serum were measured by fluorometric assay. In addition, hemodynamic parameter were also determined by 57Co-labelled microsphere technique. Results Intensity of NOS staining in the esophageal epithelium of cirrhotic rats was significantly sironger than that in normal rats. There was a NOS-positive staining area in the endothelia of esophageal submucosal vessels of cirrhotic rats, but the NOS staining was negative in controls; NO concentration of esophageal tissues homogenate or serum in cirrhotic rats were significantly higher in comparison with that of controls(P<0.01,respectively): All cirrhotic rats had portal hypertension and hyperdynamic circulatory status. Conclusion NO may play an important role in development of esophageal varices and in changes of hemodynamic pattern in cirrhosis. [
