辛伐他汀对大鼠离体心脏左心室功能的影响

目的　鉴于辛伐他汀在冠心病预防中的作用 ,探讨其对离体心脏功能的直接影响。方法　SD大鼠离体心脏 ,Langendorff灌流 ,分别给予 1,3,10 ,30和 10 0 μmol·L- 1的辛伐他汀 ,观察其 6 0min内对心脏左心室舒缩功能的影响。结果　辛伐他汀 3～30 μmol·L- 1改善心脏的舒缩功能 ,增加左心室发展压和±dp/dtmax,10～ 30 μmol·L- 1增加冠脉流量 ,30μmol·L- 1可减慢心率。当浓度达到 10 0 μmol·L- 1时 ,心脏舒缩功能明显受损 ,短时间出现心脏死亡。结论　较低浓度的辛伐他汀可以直接增强心脏舒缩功能 ,但高浓度时可损害心功能

Effects of simvastatin on contractile function in isolated rat heart

AIM Considering wide use of simvastatin in prevention of coronary artery atherosclerotic heart disease, the direct effects of simvastatin on the isolated rat hearts was investigated. METHODS The isolated rat hearts were perfused with Krebs-Henseleit solution and administrated simvastatin 1, 3, 10, 30 or 100 μmol&;#8226;L^-1, respectively, then the indexes of left ventricular function (left ventricular development pressure, ±dp/dt_max) were observed and analyzed. RESULTS The contractile function of the isolated rat heart was improved when 3-30 μmol&;#8226;L^-1 simvastatin were administrated with the decrease in heart rates at 30 μmol&;#8226;L^-1. The coronary flows were significantly increased when 10 or 30 μmol&;#8226;L^-1 simvastatin were given. While obvious damage of contractile function, even sudden death, occured when the agent was given at 100 μmol&;#8226;L^-1 dosage. CONCLUSION Proper use of simvastatin produces positive effect on the contractile function in normal rat hearts directly, but overdose of simvastatin deteriorates the contractile function.
