应用原位杂交检测乳腺癌中大炎肽基因的表达

摘    要:目的:探讨生物素标记的寡核苷酸探针,检测乳腺癌石蜡切片中大炎肽(daintain)基因的应用价值。方法:应用原位核酸杂交(ISH)和免疫组化(IHC)技术进行检测,并进行比较。结果:230例乳腺癌石蜡切片中,ISH检出的大炎肽基因阳性率为55%。ISH法检测大炎肽基因阳性率高于IHC法测出的大炎肽阳性率(53%)。大炎肽原位杂交信号主要出现在乳腺癌细胞核周边及其内外,相比之下,胞质区域的原位杂交信号较弱。原位杂交阳性率随乳腺癌淋巴结转移数目的增多而增高。结论:生物素标记的大炎肽寡核苷酸探针作为一种分子检测技术,可以较为理想地检测出乳腺癌组织中的大炎肽基因表达。

Detection of daintain mRNA expression in paraffin - embedded sections of breast carcinoma by RNA nucleic acid hybridization in situ

Abstract:Objective: Biotin - labelled oligonucleotide probes were prepared by direct chemical synthesis to detect the expression of daintain in paraffin - embedded sections of breast carcinomas. Methods: Total of 230 cases were examined by nucleic acid hybridizayion in situ(ISH) and immunohistochemistry assay(IHC). Results : The positive rate of ISH for daintain gene was 55%. The positive rate of daintain gene by ISH was higher than those of daintain monoantibody by IHC which were 55% and 53% respectively. Hybridization signals in breast carcinoma cells distributed in the both sides of the nuclei ; mainly located in the side with more cytoplasm ; in nuclei or protruded into the nucle. The hybridization signal of daintain was positive correlated with the lymphatic metastasis of breast cancer. Conclusion: Biotin- labelled daintain oligonucleotide probes and ISH could be an excellent molecular detection technique of daintain gene in paraffin - embedded sections of breast carcinomas.
