华中五味子酮对阿尔茨海默病模型大鼠学习记忆的影响及其机制的研究

摘    要:目的观察华中五味子酮(schisandrone)对阿尔茨海默病(AD)模型大鼠学习记忆及大脑内过氧化氢酶(CAT)、谷胱苷肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)活性的影响,探讨华中五味子酮对AD可能的防治作用。方法30只雄性SD大鼠,周龄8~12周,体质量250~300 g,随机分为3组:空白对照组、AD模型组和华中五味子酮干预组(每组10只)。采用侧脑室立体定向注射Aβ25-35(溶于无菌生理盐水,浓度10 mmol/L)的方法,建立AD的动物模型,并用华中五味子酮(溶于玉米油,浓度2 mmol/L)灌胃进行药物干预,测定各组Morris水迷宫逃避潜伏期,通过化学比色法观察各组大鼠脑组织CAT、GSH-Px、SOD的活性。结果与对照组比较,AD模型组大鼠的逃避潜伏期较长,而且其脑组织内CAT、SOD及GSH-Px活性较低,而用华中五味子酮干预的AD样大鼠的逃避潜伏期较AD模型组缩短(P<0.05),而且其大脑内CAT、SOD及GSH-Px活性较AD样大鼠显著增高(P<0.05或P<0.01)。结论五味子酮能够显著提高Aβ25-35介导的AD模型大鼠的学习记忆能力,可能与其提高大脑内抗氧化酶系统(CAT、SOD、GSH-Px)活性,促进氧自由基的清除有关。

Abstract:Objective To observe the effects of schisandrone on learning and memory abilities and CAT,SOD,GSH-Px activity in the brain of rats with Alzheimer-like disease and to investigate the probable preventive and therapeutic effect of schisandrone on AD.Methods Thirty male SD rats of 8~12 weeks old,250~300 g weight were divided randomly into three groups: blank control group,AD model group and schisandrone intervention group (n=10).The animal model of AD was established by 10 mmol/L Aβ_(25-35) 5 μL injection into lateral cerebral ventricle of rats and the rats of schisandrone intervention group were administrated with corn oil with 2 mmol/L schisandrone.After that,the escape latency of the rats tested in Morris water maze was recorded,the activity of CAT,SOD and GSH-Px in the brain were detected by chemical colorimetric method.Results Rats in AD group has a significant longer escape latency and a lower CAT、SOD and GSH-Px activity (P<0.05).Compared with the rats in model group,those receiving schisandrone treatment manifested decreased escape latency(P<0.05),with enhanced CAT,SOD and GSH-Px activity(P<0.05 or P<0.01) in the brain tissue.Conclusion Schisandrone can remarkably enhance the learning and memory capacity in rats with Aβ_(25-35)-induced dementia,presumably in relation to their actions to raise the activity of antioxidase system and improve the scavenging of the ROS.
