摘    要:目的:观察并探讨p16、cyclin D1基因在原发性肺癌组织中的表达水平及其与肿瘤病理生物学行为等的关系.方法:采用S-P免疫组化方法检测68例原发性肺癌标本中p16、cyclin D1蛋白的表达.结果:原发性肺癌中p16蛋白表达阳性率显著低于癌旁组织(P<0.05);cyclin D1在原发肺癌中的阳性率显著高于癌旁组织(P<0.05).二者表达与肿瘤大小、淋巴结转移、病理分级及疾病分期密切相关(P<0.05),且二者表达显著负相关(P<0.05).结论:在原发型肺癌中存在抑癌基因p16编码蛋白的表达下调以及原癌基因cyclin D1表达上调,与肿瘤的进展密切相关.提示二者可能通过影响细胞周期调控在肺癌的发生发展过程中起重要作用.

Abstract:Objective: To observe and explore the expressions of p16 and cyclin D1 in primary lung cancer and the relationship between the expressions and the pathobiological behavior of primary lung cancer.Mothods: The expressions of p16 and cyclin D1 were determined in 68 cases of primary lung cancer by using immunohistochemical techniques(S-P).Results: The positive expression rate of p16 in primary lung carcer was significantly lower than that in the peritumor normal tissues(P<0.05), and that of cyclin D1 was significantly higher(P<0.05).Expressions of p16 and cyclin D1 proteins were associated with tumor size,lymph node metastasis,pathological grades,and tumor stage(P<0.05).There was a significantly negative correlation between the expression of p16 and cyclin D1(P<0.05).Conclusion: Expression of protein encoded by tumor suppressor gene p16 is down-regulated and that encoded by oncogene cyclin D1 is up-regulated in tumorigenesis of lung cancer.The expressions are tightly associated with the progression of lung cancer,which suggests that p16 and cyclin D1 may play important roles in the progression of primary lung cancer by regulating cell cycle.
