高压氧诱导脑缺血再灌注海马CA1区神经元Bcl-2表达的时程变化

摘    要:目的观察高压氧治疗脑缺血再灌注损伤海马CA1区神经元Bcl-2蛋白表达的时程变化。方法沙土鼠前脑缺血20min后再灌注1d,3d,5d,用0.25MPa的高压氧(hyperbaricoxygen,HBO)治疗(60min/d),采用LSAB免疫组化方法,检测海马CA1区神经元Bcl-2阳性神经元。结果单纯缺血各组及压力对照组海马CA1区未见Bcl-2阳性的神经元,各HBO治疗组海马CA1区可见大量Bcl-2阳性神经元。HBO治疗3d,Bcl-2阳性神经元最多(P<0.01)。结论脑缺血再灌注损伤应尽早行HBO治疗,并且疗程应充足。

Abstract:Objective To observe the time phase changes of Bcl 2 in the gerbil hippocampus CA1 following forebrain ischemia reperfusion after treatment with hyperbaric oxygen and to provide further theoretical basis for hyperbaric oxygen treatment ( HBOT ).Methods The change of the expression of protein Bcl 2 was detected in CA1 region of gerbil hippocampus following HBOT on ischemia reperfusion using the method of labelled streptavidin biotin (LSAB) immunohistochemistry staining. For this purpose gerbils were exposed to the 0.25 MPa HBO 60 min every day for 1,3 or 5 days respectively.Results There was no expression of protein Bcl 2 in hippocampus CA1 in ischemia reperfusion groups or in hyperbaric air control groups. The expression of Bcl 2 was detected in HBOT groups and the expression of Bcl 2 in 3 d group was significantly increased compared with those in other groups (P< 0.01).Conclusion HBOT must be used earlier and longer enough to protect neurons from injury of ischemia reperfusion.
