Creatinine determination in the urine of alcaptonuric patients

Abstract:If the action of homogentisic acid is neglected, the values of creatinine determined in alcaptonuric urines by Jaffe's reaction are about 50% in excess on the average. Based on these results, misleading clinical conclusions about renal functional state can be drawn.Homogentisic acid interference can be completely avoided by previously isolating creatinine on strongly acid cation-exchanger. A modification of the Teger-Nilsson method for creatinine determination is presented. Optimum conditions for creatinine isolation on the ion-exchange column and the photometric behaviour of the mixture of homogentisic acid and creatinine with alkaline picrate are discussed.
