哌仑西平抑制豚鼠形觉剥夺性近视的效果与机制研究

摘    要:目的观察哌仑西平滴眼液对豚鼠形觉剥夺性近视的抑制作用,并探讨其作用机制。方法健康1周龄豚鼠40只,随机分为:正常组(I)、单纯遮盖组(II)、含透明质酸钠的哌仑西平滴眼液组(III)、含氮酮的哌仑西平滴眼液组(IV)。II、III、IV组豚鼠的右眼均用半透明眼罩遮盖,III、IV组豚鼠的实验眼分别每日早晚滴用含透明质酸钠的哌仑西平滴眼液和含氮酮的哌仑西平滴眼液;7周后检测4组豚鼠的双眼屈光度、眼轴长度,免疫组化法检测视网膜酪氨酸羟化酶(TH)表达水平。结果4组实验眼间屈光度两两比较II、III组无显著性差异(t=-1.580,P>0.05),余有显著性差异(P<0.05);眼轴长度两两比较I、IV组无显著性差异(t=-0.420,P>0.05),余有显著性差异(P<0.05);视网膜TH阳性细胞数两两比较I、IV组无显著性差异(t=0.323,P>0.05),余有显著性差异(P<0.05)。TH阳性细胞数与相应屈光度呈直线相关(r=0.809,P<0.01)。结论2种哌仑西平滴眼液均能抑制豚鼠形觉剥夺性近视的发展,氮酮组作用效果显著;且均能阻止形觉剥夺性近视视网膜TH表达水平的下降。

Abstract:Objective To determine the inhibitory effects of pirenzepine on form deprivation myopia (FDM)in the guinea pig and to study its mechanism.Methods Forty one-week-old guinea pigs were randomly divided into 4 groups:I control group,II form deprivation group,III pirenzepine ophthalmic solution containing sodium hyaluronate (SH) and IV pirenzepine ophthalmic solution containing azone.FDM was established in the right eyes of group II,III and IV by placement of a translucent occluder.Deprived eyes of group III and IV were respectively given the two times pirenzepine ophthalmic solutions daily.Optical measures (refraction,axial length) were taken at the end of the experiment,and immunohistochemistry was used to analyze the expression of tyrosine hydroxylase (TH) in the retina.Results There was no significant difference in refractive error between group II and III(t=-1.580,P>0.05),axial length between group I and IV(t=-0.420,P>0.05)and TH expression between group I and IV(t=0.323,P>0.05).However,there were significant differences between the other groups in refractive error,axial length and TH expression when 4 groups compared with each other.There was a linear relationship between TH positive cells and diopter(r=0.809,P<0.01).Conclusion Both pirenzepine solutions can inhibit the development of myopia,and pirenzepine with azone do better than pirenzepine with SH.Both can significantly inhibit the decrease of TH expression.
