人重组骨形成蛋白对特殊状态下兔髁突骨折愈合影响的实验研究

摘    要:目的对兔下颌骨髁突骨折断端应用人重组骨形成蛋白(rhBMP),探讨其对在缺乏维生素D供应状态下骨折端骨折愈合的作用和愈合方式。方法选择大耳兔60只,随机分为两组,然后建立下颌骨髁突骨折模型,一组给以吸附在载体胶原上的rhBMP作用实验组,另一组为空白对照组。随后分别于1、2、3、4、5、6周处死每组各5只兔子,对两组骨折断端的愈合进行骨折愈合处肉眼、透射光、X线、苏木素-伊红(HE)染色切片、电子显微镜等观察。实验结果采用析因分析。结果运用rhBMP组不论是骨折端愈合表面,还是断端组织切片结果,均表明在愈合时间、愈合的程度上较对照组好,表现在两组骨折部X线软件灰度分析差异有统计学意义(P<0.01)。结论本实验证明骨形成蛋白对缺乏充足维生素D补充的兔下颌骨髁突骨折的愈合有促进作用。

Abstract:Objective To study the role of recombinant human bone morphogenetic protein(rhBMP) in condylar fractures in Vit D-deficient rabbits.Methods Sixty Japanese white rabbits with condylar fractures of the mandibula were randomized to receive collagen-vehicled rhBMP or be treated as controls.Consequently,every five rabbits in each group were sacrificed at weeks 1 to 6,and the fracture sites were serially studied for gross pathology,translucency,radiography,hematoxylin and eosin stained histology and electronic microscopy.Data obtained were processed for factorial analysis.Results Rabbits on rhBMP showed shorter time to and better degree of healing at both surfaces and interfaces of the fracture ends compared with the controls,which was reflected by a significant difference in software-based grayscale analysis on radiography of the two groups(P<0.01).Conclusion rhBMP was shown to accelerate the healing of condylar fractures in vitamin D deficient rabbits.
