灯盏细辛治疗脑缺血再灌注损伤的时-效关系初步研究

目的:初步探讨灯盏细辛注射液对脑缺血再灌注损伤的时-效关系,以期揭示最佳的治疗周期。方法:基于线栓法阻塞大脑中动脉致大鼠局灶性脑缺血再灌注损伤模型,在动物脑缺血后第2 h、24 h、48 h、72 h和96 h时,开展灯盏细辛注射液对模型动物体重、生化指标、神经行为学和脑梗死比率的影响研究。结果:灯盏细辛注射液(1.67～6.67ml/kg/d)能明显地减少脑梗死面积并降低脑梗死比率,亦能明显促进神经功能缺失体征的提前恢复,对MCAO模型大鼠进行性的体重减轻均有不同程度的延缓作用,该作用可能与减少T NOS介导的神经毒性及自由基、过氧化脂质的产生有关。结论:灯盏细辛注射液在动物脑缺血第48h时治疗脑缺血再灌注损伤的作用最明显,存在较明显的时-效关系,可为临床合理用药提供参考。

Preliminary study of time-effect relationship of ethnomedicine herba erigerontis to cure cerebral ischemia-reperfusion injury

Objective: To explore the time-effect relationship of Ethnomedicine Herba Erigerontis in terms of curing cerebral ischemia-reperfusion injury,thereby revealing the best timing for the treatment.Methods: Based on focal cerebral ischemia-reperfusion models in rats induced by the transient occlusion of the middle cerebral artery(MCAO) by means of fishing line,a study shall be conducted to research the effect of Herba Erigerontis injection on body weight,biochemical,neurological behavior and cerebral infarction ratio of the rats models at 2 h,24 h,48 h,72 h and 96 h respectively after their cerebral ischemia.Results: Herba Erigerontis injection(1.67～6.67ml/kg/d) can significantly reduce cerebral infarction ratio,greatly advance the recovery of neurological behavior and slow down the progressive loss of body weight to varying degrees.Those effects are probably related to the decrease in T NOS-mediated neurotoxicity and less generation of free radical,lipid peroxidation.Conclusion: The time-effect relationship of Ethnomedicine Herba Erigerontis to cure cerebral ischemia-reperfusion injury appears the most obvious at 48h,which is proven to be the best treatment timing.This conclusion can be used as a reference for the clinical rational drug use.
