冠心苏合胶囊含药血清对乳鼠心肌细胞氧化损伤的保护作用

摘    要:目的:采用血清药理学方法探讨冠心苏合胶囊对过氧化氢(H2O2)致心肌细胞过氧化损伤的保护作用。方法:血清药理学方法制备含药血清(0.8g生药/kg),体外培养原代乳鼠心肌细胞,随机分为正常对照组、空白血清对照组、H2O2损伤模型组、冠心苏合低、中、高剂量(5%,7.5%,10%)干预组,建立H2O2氧化损伤模型,以不同剂量的冠心苏合含药血清进行干预。MTT法测定细胞存活率,并检测各组心肌细胞培养上清液中乳酸脱氢酶(LDH)、丙二醛(MDA)的含量及总超氧化物歧化酶(T-SOD)及过氧化氢酶(CAT)的活性。结果:100μmol/L H2O2处理2h,原代乳鼠心肌细胞存活程度适中,模型制备的重复性较好;与H2O2单纯损伤组和空白血清对照组比较,冠心苏合低、中、高(5%,7.5%,10%)剂量干预组的心肌细胞有较高存活率(P<0.01),且呈现一定的剂量依赖性;与正常细胞相比,H2O2损伤后MDA、LDH含量明显升高,T-SOD、CAT活性降低,而冠心苏合含药血清可剂量依赖性的降低损伤细胞MDA、LDH含量,升高CAT、T-SOD活性,结果具有显著统计学差异。结论:冠心苏合胶囊能够提高H2O2诱导的原代乳鼠心肌细胞存活率;下调MDA、LDH水平,上调CAT、SOD水平,对氧化损伤的心肌细胞起保护作用

Abstract:Objective :To study the effects and mechanism of the GuanXinSuHe capsules on hydrogen peroxide(H2O2) injured cardiocytes with a serum pharmacological method.Methods: Prepared the serum containing GuanXinSuHe(0.8g /kg) with Serum Pharmacological Method.Cardiocytes derived from neonatal rats were cultured in Dulbecco Modified Eagle Medium(DMEM).The cardiocytes were injured with hydrogen peroxide,and then treated with the serum containing different concerntration of GuanXinSuHe.The survival rates of cardiocytes was measured with MTT method.The content of malondialdehyde(MDA) and lactate dehydrogenase(LDH),and the activity of total superoxide dismutase(T-SOD) and catalase(CAT) were measured.Result: The neonatal cardiocytes were injured with H2O2 for 2h,and the survival rates were decreased significantly compared with normal cells(P<0.01).The content of MDA and LDH were increased significantly,but the activity of SOD and CAT were decreased.After treated with the different concentrates of drug-containing serum,the survival rate were increased with a dose dependant manner.The content of MDA and LDH were decreased and the activity of SOD and CAT were significantly increased(P<0.01).Conclusion: GuanXinSuHe capsules can protect the H2O2 injured cardiocytes by increasing antioxidation capability.
