DDVP和残杀威对淡色库蚊非特异性酯酶及乙酰胆碱酯酶作用的研究

摘    要:目的:研讨DDVP和残杀威对淡色库蚊非特异性酯酶(NSE)及乙酰胆碱酯酶(AChE)的作用,完善蚊虫抗药性的生物化学检测方法;方法:用微量滴定板法测定单个蚊虫:NSE和AChE活力;结果:DDVP和残杀威可抑制活体淡色库蚊:NSE活力,处理72h后NSE活力基本恢复到处理前水平,DDVP对NSE的抑制作用大于残杀威;抗DDVP品系和抗残杀威品系淡色库蚊AChE对DDVP和残杀威的敏感性均较敏感品系降低,抗残杀威品系AChE不敏感性较抗DDVP品系强,残杀威对淡色库蚊。AChE的抑制作用大于DDVP;结论:测定NSE检测蚊虫抗药性时,若虫已用有机磷或氨基甲酸酯类杀虫剂处理,应在处理72h后测定,测定AChE不敏感性检测蚊虫抗药性时,可选用残杀威作为抑制剂。

Abstract:Objective: To explore the roles of DDVP and propoxur for NSE and AChE of Culex pipiens pallens and to improve biochemistry assay for determination of insectieidal resistance in mosquitoes. Methods: We tested the activity of NSE and AChE of individual mosquito with microplate assay. Results: DDVP and propoxur could inhibit the activity of NSE and AChE of Culex pipiens pallens , and the activity of NSE returned to normal level of pre-treating after treating 72 h. The sensitivities of AChE of anti-DDVP and anti-propoxur strain of Cx. pipiens pallens were lower than that of sensitive strain, the insensitivity of anti-propoxur strain being high compared with anti-DDVP strain, and the inhibition action for AChE with propoxur being larger than that of DDVP. Conclusion: The NSE should be determined after treating 72 h if mosquitoes have been treated with OP and carbamates insecticides, and propoxur should be selected as inhibit agent when testing AChE insensitivity for resistance of mosquitoes.
