Abstract:Objective It is to explore the effect of Panax Notoginseng injection on the diversity structure of intestinal flora in rats with chronic renal failure.Methods 120 male SD rats were randomly divided into normal group,model group,Panax notoginseng low-dose group,Panax notoginseng medium-dose group,Panax notoginseng high-dose group,and benazepril group,with 20 rats in each group.Except for the normal group,the other groups were all established chronic renal failure models by 5/6 nephrectomy.After successful modeling,the rats in the normal group and the model group were intraperitoneally injected with normal saline 10 mL/(kg·d),and the rats in the Panax notoginseng low-dose group,medium-dose group and high-dose groups were respectively intraperitoneally injected with Panax notoginseng injection 1 g crude drug/(kg·d),2 g crude drug/(kg·d),3 g crude drug/(kg·d),and the benazepril group was given benazepril hydrochloride 5 mg/(kg·d)by gavage,all once per day,for a total of 8 weeks.24 hours after the last intervention,abdominal aortic blood was collected to detect renal function indicators[urea nitrogen(BUN),serum creatinine(SCr)],and fecal samples of the rats in each group were collected for biodiversity analysis(αdiversity analysis,βdiversity analysis,OTU analysis).Results The levels of BUN and SCr in the normal group and each treatment group were lower than those of the model group(all P<0.05),and the Panax notoginseng high-dose group was significantly lower than the Panax notoginseng low-dose group,the medium-dose group and the benazepril group(all P<0.05).αdiversity analysis showed that the Shannon,ACE and Chao indices of the model group were significantly lower than those of the normal group(all P<0.05);the ACE and Chao indices of the Panax notoginseng low and medium dose groups were significantly higher than those of the model group(all P<0.05),and the medium-dose group was significantly higher than that of the low-dose group(all P<0.05);the Shannon,ACE,and Chao indices in the Panax notoginseng high-dose group and the benazepril group were significantly higher than those in the model group and the low-dose group and middle-dose group(P<0.05),but the indexes in the Panax notoginseng high-dose group were significantly lower than those in the benazepril group(all P<0.05).β-diversity analysis showed that the sample distances between the normal group and the Panax notoginseng low-dose group,Panax notoginseng middle-dose group,and the benazepril group were relatively close;the distribution ratio of intestinal flora in rats with chronic renal failure changed significantly at the genus level,the abundances of Lactobacillus,Lachnospira,Rumenobacter,and Ruminococcus increased significantly,and the abundance of Escherichia coli-Shigella decreased significantly in the Panax notoginseng medium-dose group and Panax notoginseng high-dose group.Conclusion Panax notoginseng injection can effectively improve the structure and diversity of intestinal flora in rats with chronic renal failure,and protect kidney function.
