拳参-413对心肌肥厚大鼠心肌电学的影响

摘    要:目的:探讨拳参-413抗心肌肥厚大鼠心律失常可能的作用机制.方法:取清洁级SD雄性大鼠32只,体质量200~260 g,随机分成 ①生理盐水对照组;②心肌肥厚模型组;③ 0.5 mmol/kg的拳参-413组;④ 1.0 mmol/kg的拳参-413组.②③④组大鼠每天背部皮下注射1.0 mg/kg异丙肾上腺素,共10天.对照组每天注射相同体积的生理盐水.10天后给药组每天给予不同浓度的拳参-413,连续7天.观察拳参-413治疗前后大鼠左心室重量指数以及乳头肌跨膜电位的变化.结果:实验结果显示,拳参-413(高、低剂量)治疗后,左心室重量指数明显降低,各组心肌肥厚大鼠右心室乳头肌动作电位时程(APD)复极化各水平均不同程度地长于正常对照大鼠,拳参-413高剂量组APD较模型组比较明显缩短(P〈0.05;P〈0.01).而去极化最大速率(Vmax)、超射值(OS)和动作电位幅度(APA)以及静息膜电位(RP)水平均无明显变化.结论:心肌肥厚大鼠右心室乳头肌动作电位时程明显延长,拳参-413能够有效的缩短APD减轻心肌肥厚,说明拳参-413具有良好的抗心肌肥厚作用.

Abstract:Objective:To investigate the potentially mechanism of Potygonum bistorta. L-413 anti HCM (hypertrophic car- diomyopathy) of rats. Method:32 SD rats weight from 200 ~ 260 g were randomly devided into (1) saline control group, (2)HCM model group, (3)Polygonum bistorta. L-413(0.5 mmol/kg) group and (4)Polygonum bistorta. L-413( 1 mmol/ kg) group. (2)(3)(4)were subcutaneous injected Iso proterenol for 10 d, while control group was subcutaneous injected sa- line as the same size. Then (2)(3)(4) groups were given different concentrations of Polygonum bistorta. L-413 for the next 7 d. Observe the change of left ventricular weight index and papillary muscle transmembrane potential in rats. Result: The experiment results showed that, after the treatment by Polygonum bistorta. L-413 high and low dosage left ventricular weight index was reduce, The papillary muscle action potential duration ( APD ) of rat with myocardial hypertrophy repo- larization levels were higher than normal control rats, and the ADP of Polygonum bistorta. L-413 (1 mmol/kg) group was reduce obviously, but the maximum rate of depolarization (Vmax) ; the level of overshoot value (OS), action potential amplitude and resting membrane potential were not change extraordinary. Conclusions:The ventricular papillary muscle action potential duration of rats with myocardial hypertrophy was significantly prolonged; Polygonum bistorta. L-413 can reduce the APD and alleviate Myocardial hypertrophy, this prove that the Polygonum bistorta. L-413 has a good anti-myo- cardial hypertrophy treatment.
