肾平颗粒对IgA肾病小鼠肾组织CD44的影响

摘    要:目的:探讨肾平颗粒治疗实验性IgA肾病的作用机制。方法:采用口服牛血清白蛋白(Ⅸ讽)及尾静脉注射葡萄球菌肠毒素B(SEB)免疫复合法诱发小鼠IgA肾病模型,随机分为正常组、模型组、洛汀新组及肾平颗粒低、中、高剂量组,共6组。观察治疗前后IgA免疫复合物在肾组织中的沉积,通过免疫组化法检测CD44在肾组织的表达。结果:正常组无IgA沉积,模型组肾小球均出现IgA沉积,而治疗组IgA在肾小球的沉积较模型组明显降低(P〈0.01),正常组仅有少量CD44表达,模型组中CD44的表达明显增强,治疗组(除肾平低剂量组)CD44在肾小球的表达明显减少,并且在肾平颗粒各治疗组中呈剂量依赖关系(P〈0.01)。结论:肾平颗粒能降低IgA肾病小鼠肾组织CD44的表达,从而延缓IgA肾病的进展。

Abstract:Objective:To study the mechanism of Chinese herbal compoud Shenpingkeli on experimental IgA Nephropathy.Methods:Male BALB/C mices were randomly divided into six groups:normal group,model group,lotensin group,Shenpingkeli low,medium and high dose group.Mouse IgA nephropathy model was established by feeding with bovine serum albumin(BSA) and injection with staphyloentero-toxin B(SEB) via teil vein.Deposition of IgA immunal complex in the renal tissue was examined before and after treatment.The expression of CD44 in the kidney was assessed by immunohistochemistry.Results:There was significant IgA immunal complex deposition in the glomerulus in model group but not in normal group.Compared with model group,the deposition of IgA immunal complex in the glomerulus was significantly reduced in treatment groups(P<0.01).The expression of CD44 was weak in the glomerulus of normal group,but was significant in model group,and was reduced in treatment groups(except for group of low dose Shenpingkeli).The reduced expression of CD44 was associated with the dose of Shenpingkeli(P<0.01).Conclusion:Shenpingkeli can reduce the expression of CD44 in the glomerulus of mice with IgA nephropathy and delayed the progression of IgA nephropathy.
