胰激肽原酶对糖尿病大鼠内层视网膜细胞间黏附分子表达的影响

摘    要:目的:研究胰激肽原酶对糖尿病大鼠视网膜内层细胞间黏附分子(intercellular adhesion molecule,ICAM)表达的影响。方法:糖尿病动物造模采用健康成年Wistar大鼠腹腔内1次性注射链脲佐菌素(streptozotocin,STZ)法。Wistar大鼠78只随机分为正常对照组和糖尿病3个月组(n=33)、糖尿病6个月组(n=33),其中糖尿病大鼠又随机分为胰激肽原酶治疗组和糖尿病对照组。到期取各组大鼠及视网膜进行血管消化铺片和免疫组化,比较分析其ICAM表达的差异。结果:正常组大鼠视网膜可见ICAM有所表达,但糖尿病大鼠ICAM表达增加,且随病程进展而增强(P=0.03)。糖尿病3个月组,胰激肽原酶治疗组ICAM较同期糖尿病对照组表达下降(P=0.036),但糖尿病6个月组间则未见明显差异。结论:糖尿病视网膜病变与ICAM异常表达有关,早期应用胰激肽原酶可能对其有一定抑制作用。

The Impact of Kininogenase on the Expression of Intercellular Adhesion Molecule (ICAM) in the Retinas of Diabetic Rats

Abstract:Objective:To investigate the effects of kininogenase on the expression of ICAM in the diabetic rats' retinas. Methods:66 streptozotocin-induced diabetic wistar male rats were divided into 2 groups randomly, kininogenase group (DMP) and diabetic control group (DMC). Rats of each group were fed for 3 or 6 months (DM3mP, DM3mC or DM6mP and DM6mC, respectively). While 12 normal wistar rats composed the healthy control group. At the end point, flat mounted retinas were digested to analysis the retinal vascular structure, and paraffin-embedded ones were used for immunohistochemical studies to analysis the differences in the expressions of ICAM between each group. Results: The expression of ICAM differs from retina to retina in the normal control group. While the ICAM levels were higher in the diabetic groups, and they were increasing with the prolonged courses of diabetes(P = 0. 03). In addition, the expression of ICAM was weaker in DM3mP group than that in DM3mC group (P = 0. 036). There was no difference between DM6mP group and DM6mC group, however. Conclusion: Abnormal expression of ICAM may be involved in the mechanism of diabetic retinopathy and kininogenase may be helpful in the treatment of the disease of its early stage by inhibiting its expression to some degree.
