石榴皮提取物对Caco-2细胞生长抑制作用实验

目的探讨石榴皮提取物对人源性结肠癌细胞Caco-2的体外抑制作用。方法培养人源性结肠癌细胞Caco-2,制作石榴皮提取物并配置成不同浓度的提取液,将这些不同浓度的石榴皮提取液加入到培养至对数生长期的Caco-2细胞中,一段时间后,用3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐(MTT)对给予不同浓度石榴皮提取物的肿瘤细胞进行染色,观察不同浓度石榴皮提取物对Caco-2细胞生长抑制程度,并计算抑制率。结果石榴皮提取物在8 g/L时对Caco-2细胞有较强的抑制作用,24、48、72 h的细胞生长抑制率分别为(50.65±5.18)%、(57.21±2.07)%、(69.88±2.14)%;在72 h时对Caco-2细胞的IC50是2.89 g/L。结论石榴皮粗提物在高浓度时具有良好的抑制Caco-2细胞增殖的作用。

Study on the Inhibitory Effect of Pomegranate Rind on the Cell Growth of Caco-2

Objective To explore the inhibiting effect in vitro of pomegranate peel extract on humanderivedcolon cancer cells Caco-2 in vitro.Methods Human colon cancer cells derived Caco-2 were cultured,the pomegranate peel extract was prepared and distributed to different concentrations,then pomegranate rind extract of different concentrations was added to the culture of Caco-2 cells in logarithmic growth phase.After some time,3-（4,5-dimethylthiazole-2）-2,5-di-diphenyl oxide bromide（MTT） was used to stain the tumor cells with different concentrations of pomegranate rind extracts,the growth inhibition effect of different concentrations of pomegranate peel extract on Caco-2 cell was observed and inhibition rates were calculated.Results The pomegranate peel extract significantly inhibited the growth of colon cancer cells Caco-2 when concentrations of pomegranate peel extract reached 8 g/L.As time prolonged,the inhibition rates of the cell growth were （50.65 ±5.18）％ in 24 h,（57.21 ±2.07）％ in 48 h and （69.88 ±2.14）％ in 72 h.The 50％ inhibiting concentration （IC50） of colon cancer cells Caco-2 was 2.89g/L in 72 h.Conclusion The inhibition effect of pomegranate peel extract？ on the growth of human-derived colon cancer cells Caco-2 is remarkable at high concentrations.
