宫颈鳞癌组织圆柱瘤基因mRNA表达及其意义

摘    要:目的探讨宫颈鳞癌组织中圆柱瘤基因(CYLD)mRNA的表达及其临床意义。方法采用原位杂交法检测45例宫颈鳞癌、64例官颈上皮内瘤变(cIN,包括CINI26例、CIN Ⅱ-Ⅲ38例)及20例正常宫颈组织中CYLD mRNA的表达,分析其阳性表达率与宫颈鳞癌临床病理特征的关系。结果CYLDmRNA在宫颈鳞癌、CINⅡ—Ⅲ、CINI及正常宫颈组织中的阳性表达率分别为42.2%、52.6%、73.1%、80.0%,宫颈鳞癌、CINⅡ-Ⅲ组织中CYLDmRNA的阳性表达率明显低于正常宫颈组织,差异均有统计学意义(x2=7.85、4.42,P〈0.05)。宫颈鳞癌CYLDmRNA表达与临床分期及淋巴结转移密切相关(x^2=4.64、5.59,P〈0.05),但与组织学分级、肿瘤大小、病人年龄无关。结论CYLDmRNA低表达可能与宫颈癌的发生、发展及转移有关。

EXPRESSION OF CYLD MRNA IN CERVICAL SQUAMOUS CELL CARCINOMA TISSUES AND ITS SIGNIFICANCE

Abstract:Objective To study the expression of CYLD mRNA in squamous cell carcinoma (SCC) of the cervix and its clinical significance. Methods In situ hybridization was used to detect the expression of CYLD mRNA in tissues of cervical SCC in 45 cases of the cancer, 64 cases of cervical intraepithelial neoplasia (ClN)-including 21 of CIN I and 38 of CIN Ⅱ-Ⅲ -and 20 cases of normal cervical tissue. The correlation between their positive expression rates and clinicopathologic characters was analyzed. Results The positive expression rates of CYLD mRNA in cervical SCC, CIN Ⅱ-Ⅲ. CIN I and normal cervical tissue were 42.2 ~, 52.6~, 73.1~/00, and 80~, respectively, that in SCC and CIN Ⅱ-Ⅲ were obviously lower than that in the normal cervical tissue, the differences were statistically significant (x 2 = 7.85,4.42 ; P〈0.05). The expression of CYLD mRNA was closely correIated with clinical stage and metastasis (x 2= 4.64, 5. 59; P 〈0.05), but not with histological grade, tumor size and patient age. Conelusion The low expression of CYLD mRNA in cervical carcinoma tissues is probably related with the occurrence, develop- ment and metastasis of this malignancy.
