清热化瘀颗粒对脑缺血预处理大鼠肌醇需求激酶1表达的影响

摘    要:目的: 观察清热化瘀颗粒对脑缺血预处理大鼠缺血再灌注后肌醇需求激酶1(IRE1)mRNA及其蛋白表达的影响。 方法: 健康雄性SD大鼠80只,随机分为4组:假手术组(SO)、脑缺血再灌注组(MCAO)、脑缺血预处理组(BIP)、清热化瘀颗粒组(QRHY),每组按再灌注后12 h,1,2,3 d 4个时间点平均分为4个亚组。制备脑缺血预处理模型,缺血预处理后给大鼠灌胃14 mL·kg-1清热化瘀颗粒溶液,连续3 d,每天上、下午各1次,MCAO组和BIP组给予等量的生理盐水。用实时荧光定量PCR和Western blot法观察再缺血后各个时间点IRE1mRNA及其蛋白的表达变化。 结果: MCAO组IRE1mRNA及其蛋白表达均于缺血再灌注后12 h开始明显上升,1 d达高峰(P < 0.01),随再灌注时间延长其表达逐渐下降,但仍保持较高表达水平(P < 0.01);BIP组较MCAO组IRE1mRNA及其蛋白表达明显升高(P < 0.05或P < 0.01),QRHY组较BIP组进一步升高其表达(P < 0.05或P < 0.01)。 结论: 脑缺血预处理能够诱导脑缺血损伤后IRE1的表达,清热化瘀颗粒可进一步促进其表达。

Abstract:Objective: To observe the effect of the Qingre Huayu granule (QRHY) on the inositol requiring enzyme 1(IRE1) mRNA and protein after focal ischemic preconditioning in rats. Method: Eighty healthy male SD rats were randomly divided into four groups:sham-operation group, middle cerebral artery occlusion (MCAO) group,brain ischemia preconditioning(BIP) group and QRHY group.Each group was further divided into 4 subgroups according to ischemia/reperfusion(I/R) duration of 12 h,1,2, 3 d.The BIP models was established and 14 mL·kg-1 QRHY granule solution was ig given twice everyday for 3 days after preconditioning. But the MCAO and BIP group was ig given same amount physiological saline.The expression of IRE1 mRNA and protein was detected by Real time PCR and Western blot. Result: The expression both of IRE1 mRNA and its protein all increased and reached the peak at 24 h, then decreased continuously(P<0.05). BIP could increase their expression (P<0.05 or P<0.01). QRHY treatment could further induce its expression compared with BIP group(P<0.05 or P<0.01). Conclusion: BIP could upregulate the expression of IRE1 after focal ischemic injury in rats. QRHY could augment the expression of IRE1 induced by BIP.
