木犀草素与牛血清白蛋白相互作用的荧光光谱法研究

摘    要:本文首次采用荧光光谱法研究传统中药白毛夏枯草中活性成分木犀草素与牛血清白蛋白(BSA)的猝灭机制。实验研究了木犀草素在291 k和310 k条件下与BSA的猝灭情况,通过计算求得不同温度下的热力学参数。结果表明木犀草素对牛血清白蛋白荧光的猝灭机制属于静态猝灭过程,二者之间的作用力主要为范德华力和氢键。此外,实验还考察了木犀草素对BSA构象的影响。

Abstract:The interaction between luteolin,which was the active constituent of the traditional Chinese medicine Decumbent bugle herb,with bovine serum albumin(BSA) had been studied by fluorescence spectrometry for the first time.In this paper,the interaction between luteolin with BSA were investigated at 291K and 310K by fluorescence spectroscopy,respectively.The thermodynamic parameters under different temperatures were calculated.The data obtained from fluorescence quenching experiments showed that luteolin was bound to BSA and the quenching type was static quenching.The results indicated that the interactions between luteolin with BSA were driven mainly van der Waals forces and hydrogen bonds.Besides,the interaction of BSA with luteolin in the conformation of BSA was also investigated.
