摘    要:目的 探讨牛磺酸对大鼠肝脏缺血再灌流损伤的作用及机制.方法 48只实验大鼠随机分为三组:假手术组(A组)、缺血-再灌流组(B组)和给予牛磺酸后缺血-再灌流组(C组).检测再灌流30 min的肝组织钙含量、丙二醛(MDA)含量和谷胱甘肽过氧化物酶(GSH-PX)活力.并同步观察肝组织光镜和电镜下的形态变化. 结果 B组肝组织钙含量和MDA含量明显高于A组(P<0.01),而GSH-PX活力低于A组(P<0.01).C组肝组织钙和MDA含量低于B组(P<0.01, P<0.05),而GSH-PX活力高于B组(P<0.05).肝损伤病理改变C组轻于B组(P<0.05).结论 牛磺酸能显著减轻大鼠肝脏缺血再灌流损伤,其机制与减轻钙超载、抑制氧自由基生成和增强氧自由基清除有关.

Abstract:Objective To explore the effects and mechanism of taurine on ischemia-reperfusion(I/R)injury in rat livers.Methods The rats were divided into sham operation group(group A),ischemia-reperfusion group(group B)and group undergoing I/R after taurine administration(group C).The contents of calcium,methane dicarboxylic aldehyde(MDA)and activity of glutathione peroxidase (GSH-P_X)in the livers were detected at 30 minutes after reperfusion.The morphological changes of the livers under optical and electric microscopes were simultaneously observed.Results Compared with group A,the contents of calcium and MDA in the group B were markedly higher( P <0.01),while the activity of GSH-P_X was weaker ( P <0.01).Compared with group B,group C had a lower contents of calcium and MDA ( P <0.01, P <0.05),but a stronger activity of GSH-P_X( P <0.05).Pathological changes in the group C were less severe than those in group B ( P <0.05).Conclusion Taurine can significantly relieve the injury induced by I/R in rat livers.The mechanisms may be connected with the reduction of calcium overload,the inhibition of oxygen free radicals production and the enhancement of cleaning these free radicals.
