摘    要:目的探讨吲哚-3-甲醇(13C)对人肝癌细胞株SMMC.7721的增殖和凋亡的影响。方法不同浓度的13C(100、150、200、250、300、350Ixmol/L)处理细胞48h后,显微镜下观察细胞生长状况;采用WST-1法检测细胞增殖情况,Hoehest33258染色、TUNEL染色检测细胞凋亡情况,Westernblot法检测凋亡相关蛋白。结果13C能够抑制SMMC.7721细胞增殖,诱导细胞凋亡,处理浓度在250μmol/L以下时,细胞生长变慢;处理浓度达到300μmol/L时,大量细胞凋亡。13C浓度高于200μmol/L时CytC、Cleavedcaspase.9和Cleavedcaspase.3蛋白表达明显增加,且随着13C浓度的增加三种蛋白的表达也明显增加。结论13C在体外实验条件下可抑制人肝癌细胞株SMMC.7721增殖并诱导其发生凋亡。

Abstract:Objective To investigate the effects of indole-3-carbinol on the proliferation and apoptosis of human liver cancer cell line SMMC-7721. Methods The growth status of SMMC-7721 was observed under Microscope. The inhibition of cell proliferation was assayed by WST-1 agent, and the apoptosis was shown with Hochest33258 and TUNEL staining. The apoptosis related protein were detected by Western blot. Results I3C could inhibit the proliferation of SMMC-7721 and induced the apoptosis in a dose-dependent manner. The cell proliferation was slow at doses below 250 μmol/L of I3C. And a large proportion of apoptotic cells were observed at doses above 300 μmol/L of I3C. Furthermore , the expressionsof Cyt C, Cleaved caspase-9 and Cleaved caspase-3 increased significantly when I3C at concentrations higher than 200μ mol/L.With the increase of I3C concentration, three kinds of protein expression also increased significantly. Conclusion I3C could significantly inhibit the human liver cancer cell proliferation and induce apoptosis in a dose dependent manner.
