乳腺癌患者外周血浆中RAR-β_2基因启动子异常甲基化分析

目的:探讨RAR-β2基因(retinoic ac id receptorβ2 gene)启动子在散发性乳腺癌患者血浆中异常甲基化状况。方法:用甲基化特异PCR(MSP)法和实时荧光定量PCR法对62例散发性乳腺癌患者血浆和15例良性乳腺增生患者血浆中的RAR-β2基因启动子异常甲基化状况进行检测。结果:散发性乳腺癌患者血清中37%(28/62)发生了RAR-β2基因启动子甲基化,良性乳腺增生患者中均未检测到RAR-β2基因的甲基化。RAR-β2基因启动子甲基化状态与淋巴结转移和临床分期有相关性(P<0.05)。结论:乳腺癌患者外周血浆中RAR-β2基因启动子异常甲基化是散发性乳腺癌发生的早期事件,有望成为乳腺癌早期诊断的指标。

Analysis of Retinoic Acid Receptor β_2 Promoter in Blood Plasma of Patients with Breast Cancer

Objective To investigate the abnormal methylation status of retinoic acid receptor β2 promoter in the plasma of patients with sporadic breast cancer.Methods Sample of 62 sporadic breast cancer and 15 benign breast tumor were collected and determined with methylation-specific PCR(MSP) and real-time quantitive PCR.Results The methylation rates of RAR-β2 gene promoter were observed in the plasma of 37%(28/62) patients with breast cancer,but no methylation was determined in the plasma of patients with benign proliferative breast tumor.The methylation status of RAR-β2 promoter were significantly associated with the lymph node metastasis and clinical typing.ConclusionThe methylation of RAR-β2 promoter was the early event,which could be an index for early diagnosing the breast cancer.
