腹膜透析液对鼠腹腔巨噬细胞功能的影响

目的 探讨含不同浓度葡萄糖的腹膜透析液不同时间作用后对巨噬细胞 (Mφ)功能的影响。方法取经3%硫乙醇酸盐培养基腹腔注射诱生的小鼠腹腔Mφ 在体外培养条件下 ,用含不同浓度 (1.50%、2.50%和4.25% )葡萄糖的腹膜透析液分别作用10、30和60min后 ,检测Mφ 吞噬功能、一氧化氮 (NO)产量和对四甲基偶氮唑盐 (MTT)还原能力。结果 观察组Mφ 吞噬功能、NO产量和MTT还原能力均明显低于对照组 (均P<0.01) ,且葡萄糖浓度越高、作用时间越长 ,各项指标下降越明显。结论 短时间腹膜透析液作用即可损害巨噬细胞活力及其他功能 ,这种影响与腹膜透析液葡萄糖浓度和作用时间长短有关

Influence of peritoneal dialysate on immune function of macrophage in mice

Objective To investigate the influence of peritoneal dialysate on immune function of macrophage. Methods Cultured mouse peritoneal macrophages were exposed to peritoneal dialysate containing different concentration of glucose(1.50%, 2.50% and 4.25%)for 10, 30 and 60min, then, their phagocytic function, nitric oxide(NO) production and MTT reduction ability were measured.Results The phagocytic function, nitric oxide(NO) production and MTT reduction ability of experimental groups were remarkably lower than those of control,and the functions in 4.25% glucose-containing dialysate group and exposure for 60min group were reduced most. Conclusion Short-time dialysate exposure may have detrimental effects on viability and other function of macrophage, and these effects may be correlated with concentration of glucose in dialysate and exposure time.
