液相色谱法和超高压液相色谱-串联质谱法测定太湖水中微囊藻毒素的比较

摘    要:目的:比较高效液相色谱法和超高压液相色谱-串联质谱法(UPLC-MS/MS)两种方法,分别考察其监测太湖水中MC-LR、RR、YR三种微囊藻毒素的优缺点。方法:分别以HPLC法和UPLC-MS/MS法进行测定,用回归法考察两种方法测定结果的相关性,比较测定结果。结果:两种方法的定量结果在统计学上无显著性差异,具有良好的相关性(P>0.05);对HPLC法方法定量限水平(RR、LR、YR方法定量限分别为0.011μg/L、0.015μg/L、0.017μg/L)以上的样品,两种方法的检出率基本相符;方法定量限水平以下的样品,由于UPLC-MS/MS具有更高的灵敏度,检出率高于HPLC法。结论:两种方法均能够应用于太湖水中微囊藻毒素的测定。HPLC法因其准确、实用和配置成本相对低的优点,更适合推广,可用于太湖水中微囊藻毒素的常规监测。UPLC-MS/MS方法灵敏度高,但配置成本高,更适用确认验证。

Abstract:Objective: To compare two methods(HPLC and ultra-performance liquid chromatography with mass spectrometers(UPLC-MS/MS)) for detecting three MCs(The MC-LR,RR and YR)in Taihu Lake.Methods: Determination was done by HPLC and UPLC-MS/MS separately,and regressive method was adopted to observe the correlation of the results of the two methods.Results: The test demonstrated that there was no remarkable difference between the results of the two methods,with good correlations(P>0.05);When the samples′ concentration was higher than LOQs of HPLC(LOQ of RR,LR,YR were 0.011 μg/L,0.015 μg/L,0.017 μg/L),the detection rates were similar between the two methods.When under the LOQs,the detection rates of UPLC-MS/MS were higher for it′s more sensitive.Conclusion: Both methods can be used for determination of MCs in Taihu Lake.HPLC is more suitable to popularize for its reliability,practicality and less equipment cost,which can be used for the routine monitoring.
