地黄多糖诱导骨髓间充质干细胞分化为神经细胞最佳浓度探索

摘    要:目的探讨地黄多糖对骨髓间充质干细胞(MSCs)诱导分化为神经细胞的最佳浓度。方法利用贴壁法分离纯化MSCs,不同浓度地黄多糖诱导其向神经细胞分化,β-巯基乙醇诱导作为对照,光镜下观察细胞形态,采用免疫细胞化学方法计算神经元和胶质细胞阳性数。结果不同浓度地黄多糖诱导培养24h后均可表现为典型的神经细胞样形态,但以中浓度地黄多糖组神经细胞数最多,且神经元比例高于其他两组。结论地黄多糖诱导MSCs分化为神经元以中浓度最佳。

Abstract:Objective:To probe the optimun concentration of rchmannia glutnosa polysaccharides(RPS)inducing bone marrow mesenchymal stem cells(MSCs) differentiated into neurons in rats.Methods:MSCs were obtained from thighbones marrow of rats and expended in culture medium.Then MSCs were induced by se concentration RPS in serum-free medium at different period.The morphological changes of the cells were observed under phase contrast microscope.The numbers of neuron and astrocyte were evaluated by immunocytochemisty staining.Results:After induction of 24h,the differentiated cells showed typical neural morphology in all groups,but the numbers of neuron and astrocyte in meddle concentration group was more than the other groups,and the rate of neuron more than the others. Conclusion:10% PRS was the best concentration induced MSDCs differentiated into neurons.
