G6PD缺乏时红细胞膜磷脂脂肪酸组成的改变

摘    要:探讨G6PD缺乏时红细胞膜磷脂的氧化性损伤用氯仿:甲醇(2∶1V/V)抽提膜脂质,通过硅胶柱层析分离得到磷脂,经薄层层析分离得磷脂酰乙醇胺(PE)、磷脂酰丝氨酸(PS)和磷脂酰胆碱(PC)采用TLC扫描仪测定各种磷脂的含量用甲醇提取PE,PS及PC,经快速酯交换得到脂肪酸甲酯、采用毛细管气相色谱分析各种磷脂中脂肪酸的组成情况结果:G6PD缺乏时,PS含量下降,红细胞膜总磷脂及PS中不饱和脂肪酸(C18∶1,C18∶2)的含量均减少,PE中C16∶0的含量也减少实验结果推测,G6PD缺乏时红细胞膜磷脂改变,PS含量的减少及PS中不饱和脂肪酸含量的下降可能与氧化性损伤有关

Abstract:Studied was the oxidative damage of eythrocyte membrane phosphalipid in G6PD deficiency. With chloroform and methanol (2:1 V/V ), lipid was extracted through silica-gel column chromatography and further decomposed into PE, PS and PC, which were then measured by TLC screener. Methanol was used to extract PE, PS and PC, from which fatty acid methyl ester was got through rapid lipid exchange. With capillary collumn gas chromatography the structure of fatty acid was analyzed in the phosphalipid. Result: G6PD deficiency witnessed the decrease of PS content; the contents of unsaturated fatty acids (C 18 :1, C 18 :2) were both reduced in total phosphalipids and PS; and the content of C 16 :0 fell in PE. From the experimental consequence it might be inferred that when G6PD is short, the oxidative damage may have a bearing on the phosphalipid of eythrocyte membrane, the loss of PS content and the content drop of unsaturated fatty acid.
