| 摘 要： | 目的:观察表没食子儿茶素没食子酸酯(EGCG)对人肾小管上皮细胞(HK-2)结缔组织生长因子(CTGF)mRNA和蛋白表达的影响并探讨其可能的机制。方法:体外培养HK-2细胞,EGCG分别以25μmol/L、50μmol/L、100μmol/L作用于体外培养的HK-2细胞48h。用ELISA法和RT-PCR检测HK-2细胞CTGF蛋白含量及mRNA表达水平。结果:与对照组比较,EGCG组HK-2细胞CTGF表达显著下降(P<0.01),且呈浓度依赖关系。结论:EGCG能抑制HK-2细胞CTGF的表达。

| Abstract: | Objective：To explore the effect of EGCG on the mRNR and protein expression of connective tissue growth factor （CT- GF） in HK 2 cells and the probable mechanism. Methods： HK-2 cells were cultured and incubated with various concentration （25umol/L, 50umol/L, 100umol/L） of EGCG for 48h. RT-PCR and EIASA were employed to examine the follows： the mRNA and protein expression of CTGF. Results：Compared with the control group, the expression of CTGF in EGCG group were significantly lower at 48h（P〈0.01）. Conclusions： EGCG can up-regulate the expression of CTGF in HK-2 cells. |
