黑素瘤细胞体外穿透性体内转移性及tPA释放量研究

摘    要:tPA是蛋白分解酶类,在肿瘤细胞侵袭和转移中起重要作用。黑素瘤细胞株可释放tPA。我们研究了A_(375)M和A_(375)P两株细胞株的体外穿透性,A_(375)M对基底膜的穿透能力是A_(375)P的4.3倍,体内的转移性前者较后者高,tPA释放量前者多于后者,表明tPA产量和肿瘤细胞的转移性相关。

Abstract:Tissue plasminogen activator(tPA)can be produced bymelanoma cells.tPA,a class of proteolytic enzymes,are thought to playan integral role in invasion and metastasis of cancer cells.The invasive andmetastatic potential of A_(375M)and A_(375P)were studied in vitro and in vivo.In the in vitro invasion assay,4.3 fold A_(375M)cells penetrated a syntheticextracellular membrane barrier than A_(375P).When injected into nude mice,A_(375M)produced metastases more frequently than A_(375P).The more aggres-sive A_(375M)cell line secreted more tPA than the relatively indodent A_(375P)cell line.These research provide further evidence that tPA production maybe related to the metastatic capacity of aggressive tumor cells.
