CEA重组痘苗病毒对实验性CEA阳性肝癌的预防和治疗作用

摘    要:目的:探讨CEA重组痘苗病毒(CEA—rV)对小鼠CEA阳性肿瘤的预防和治疗作用。方法:①预防作用观察:将实验小鼠分为4组,第1、2组和第3、4组分别皮下接种野生型痘苗病毒(W—VV)和CEA—rV3次,末次接种后,第1、3组皮下注射同源鼠CEA阴性肝癌细胞(Hepa—CEA),第2,4组接种Hepa—CEA^ 。肿瘤细胞接种后每周测量肿瘤体积,共6周。②治疗作用观察:第1、2组先皮下注射Hepa—CEA^-,3、4组接种Hepa-CEA^ ,7d后1、3组皮下接种W—VV,2,4组接种CEA—rV。肿瘤细胞接种后每周测肿瘤体积,共6周。结果:①预防作用:接种CEA—rV和Hepa—CEA^ 组(第4组)小鼠在观察期内无肿瘤形成,其他小鼠皮下均有肿瘤形成,并且以相近速度增长。②治疗作用:接种Hepa—CEA^ 和CEA—rV组(第4组)小鼠皮下肿瘤生长缓慢,瘤块较小,其他3组肿瘤生长迅速,瘤块较大。整个实验过程中各组小鼠均未表现出其他任何不良反应。结论:CEA—rV对实验性CEA阳性肿瘤具有良好的预防和治疗作用,而且预防作用更佳。

Abstract:Aim: To study the preventive and therapeutic effect of CEA recombinant vaccinia virus (CEA-rV) on CEA positive tumor. Methods: Observation of preventive effect: 40 mice were divided into 4 groups.The 1st and the 2nd groups were vaccinated by subcutaneous injection (SI) with wild-type vaccinia virus (W GAAB2 VV) and the 3rd and the 4th groups were vaccinated with CEA-rV for 3 times,respectively.After final vaccination, the 1st and the 3rd groups were injected with Hepa-CEA - tumor cells and the 2nd and the 4th groups, with Hepa-CEA + tumor cells.Obsesvetion of therapeutic effect: 40 mice were divided into 4 groups. The 1st and the 2nd groups were SI with Hepa-CEA - tumor cells, and the 3rd and the 4th groups, with Hepa-CEA + tumor cells. After 7 days, the 1st and the 3rd groups were injected with W-VV and the 2nd and the 4th groups,with CEA GAAB2 rV. The tumor bulk was observed once a week after the injection of tumor cells. Results: Preventive effect: subcutaneous tumor grew rapidly at approximately same rate (P> 0.05) in the 1st, 2nd, and 3rd groups, however, no tumor grew in the mice of the 4th group in which CEA GAAB2 rV and Hepa-CEA + tumor cells were injected; observation of therapeutic effect: the subcutaneous tumor were observed in the 1st, 2nd, and 3rd groups and the tumors grew relatively fast, no statistic significance between these 3 groups was obtained (P>0.05), whereas the tumor in the 4th group, which were injected with Hepa-CEA + tumor cells and CEA GAAB2 rV, grew slowly and the tumor mass was smaller. The statistic difference was significant in comparison with the other 3 groups (P< 0.01). During the whole experimental period, no obvious toxic or side-effects were observed.Conclusion: It is indicated that CEA-rV has well preventive and therapeutic effect on CEA + tumor in mice, and the preventive effect is better than therapeutic effect.
