养心通脉有效部位方对心肌缺血大鼠心血管内皮细胞血管生成的影响

摘    要:目的:探讨养心通脉有效成分部位方(YTⅡ)促血管生成的作用靶点及其机理。方法:设YTⅡ组和各对照组养心通脉原方(YTⅠ)组、麝香保心丸(SBW)组、重组牛碱性成纤维细胞生长因子(bFGF)组、空白血清组、生理盐水(NS)组].并进行心肌缺血模型大鼠心脏血管内皮细胞的培养;观察比较在血管生成的过程中YTⅡ组及各对照组含药血清对心脏血管内皮细胞“增殖”、“迁移”和“管腔结构形成”不同环节的干预作用。结果:各组药物干预后,心肌缺血大鼠冠状血管内皮细胞的增殖率、迁移率、成管率3类指标均呈现出bFGF组〉YTⅡ组〉YTI组〉SBW组〉NS组〉空白血清组的递减趋势。YTⅡ和bFGF对血管生成的不同环节均有明显的促进作用,且显著优于其他各组(P均〈0.01),但YTⅡ组和bFGF组无显著差异(P〉0.05)。结论:养心通脉有效部位方益气活血,能刺激血管内皮细胞“增殖”、“迁移”和“管腔结构形成”不同环节,促进缺血心肌的血管生成。

Abstract:Objective: To investigate the impact and mechanism of the effective components recipe of Yangxintongmai Formula (YT-Ⅱ ) on angiogenesis of endothelium of rat with myocardial ischemia. Methods: The endothelial cell derived from the myocardial ischemic rat model was cultured and divided into YT-Ⅱ group and controlled groups including Yangxintongmai formula (YT Ⅰ) group, Shexiang-baoxin wan (SBW) group, b--FGF group, normal saline (NS) group and blank control group. Then the interference action on the "proliferation", "migration" and "lumen formation" of endothelial ceils in every group was observed and compared. Results: The YT-- Ⅱand b--FGF can im- prove the process of angiogenesis significantly, and better than other groups (P〈0.01 all). The proliferation rate, migration rate and lumen formation rate presented with the trend of b--FGF group〉 YT- Ⅱ group〉 YT-Ⅰ group〉 SBW group〉NS group〉blank control group; There was no significant difference between YT-Ⅱ group and b--FGF group (P〉0.05). Conclusion: The results indicated that YT-Ⅱ can more stimulate "proliferation", "migration" and "lumen formation" of endothelial cells, improve the angiogenesis of ischemie myocardium.
