| 摘 要： | 用1高碘酸处理结肠癌组织,然后免疫小鼠制备单克隆抗体.获得2株产生高效价、针对结肠癌抗原的抗体杂交瘤细胞株C_8和C_(102)。经免疫组织化学染色后知其抗原定位于结肠癌腺体的上皮细胞及其分秘物.由Immunoblotting知其抗原的分子量为20万道尔顿.初步研究表明,C_8和C_(102)是特异性较强的针对结肠癌的单克隆抗体(IgG2b).经高碘酸(或胰酶)处理证明,其相应抗原决定基部分是蛋白质而不是多糖.

| Abstract: | Human colon carcinoma tissue' wastreated with 1% periodic acid andthen it was injected to BALB/c miceand the monoclonal antibodies wereraised.Two hybridoma cell lines C_8and C_(102) which can secrete hightiter of McAb to colon carcinomaAg were established.The Ag wasfound to be located in the epithelialcells of colon carcinoma glands andtheir secretions by using immuno-histochemical stain.The molecularweight of the Ag is 200KD by immu-noblotting.It was suggested by pre-liminary study that C_8 and C_(102)might be specific McAb(IgG2b)tocolon carcinoma.The study conductedby means of treatment with periodicacid or trypsin showed that thedeterminants of the Ag are not car-bonhydrate but protein.These anti-bodies directly to protein are usefulfor screen and isolation of the GENEof colon carcinoma. |
