羊栖菜多糖诱导肿瘤细胞凋亡的研究

摘    要:目的 :揭示羊栖菜多糖 (SFPS)的抗肿瘤作用机制。方法 :采用流式细胞仪观察羊栖菜多糖对肿瘤细胞周期及细胞凋亡的影响。采用Fluo-3/AM探针标记 ,激光共聚焦技术观测细胞内 [Ca2+ ]i。结果 :羊栖菜多可阻滞SGC-7901人胃癌细胞由G0 /G1期进入S期 ,升高细胞凋亡指数 (APO% )。可使SGC-790 1细胞内 [Ca2+ ]i先升高然后下降 ,给CaCl2 后 ,[Ca2+ ]i又升高。结论 :羊栖菜多糖通过升高肿瘤细胞内 [Ca2+ ]i启动肿瘤细胞凋亡机制而达到抗肿瘤的作用 ,[Ca2+ ]i升高时Ca2+ 来源于细胞内钙库释放。

Abstract:Objective: To study mechanism of antitumor a ctivity of Sargassum Fusiforme Polysaccharide(SFPS). Method: The effect on cell cycle and apoptosis was studied with flow cytometry(FCM). Intracellular calcium concentration [Ca2+ ]i was marked with Fluo-3/AM and measured with laser scanner confocal microscope(LSCM). Result: SFPS inhibited G 0/G1 stage SGC-7901 from entering to S stage an d increased APO%. The [Ca 2+ ]i showed a transient rise and return to th e original level. The concentration could be raised again by administering CaCl2. Conclusion:The antitumor effect of SFPS seems to be accomplished through the apoptosis associated with the increase in intracellular calcium concentration. Intracellular stores release the calcium during its acti on.
