大鼠窦房结中央区细胞类型的形态特征和数量分析

摘    要:目的:为研究起搏机需辨认,计数和分离大鼠窦房起搏(P)细胞,方法:根据最早起搏点和细胞电位记录确定大鼠窦房结中央区位置,取窦房结连续切片,HE和Masson染色,以光镜,图像仪及电镜辨认,分析窦房结内各类细胞形态学特点和各类细胞数量,结果,从细胞大小,外形,染色浓淡,核/胞浆比,核面积和核灰度,肌原纤维分化程度等指标,可区分各类细胞,并算出各类细胞的数量比,结论:确立了各类细胞的辨别标准,中央区P

Abstract:Objective:To recognize, count and isolate pacemaking(P) cells from the central region of rat SAN in study of pacemaking mechanism. Methods: On the basis of observation of the earliest pacemaking point and the recording of cell action potential, the position of rat SAN was determinated. After continuous slices, HE or Masson stain was done, Different cell types were identified and counted by light microscopy, image analysis system, and electron microscopy. Re-sults; P cell could be distinguished from transitional(T) cells, fibrocytes and neurogliocytes, ac-cording to the indexes of cell size, cell shape, stain depth, ratio of neucleus and cytoplasm area, gray of neucleus and myofibril mature grade, and the ratio of different cell types in the central re-gion was counted. Conclusion: Morphological features of P cell and other cells in rat SAN have been established. P cell number in the central region of rat SAN was between 1200 and 2000, ac-counting for 13. 1 % of the total numbers of cells. The percentage of T cells was 44%, and fibro-cytes 17. 1%.
