电针天枢穴和太冲穴对便秘型肠易激综合征模型小鼠5-羟色胺信号系统的影响

摘    要:目的:观察电针天枢穴和太冲穴对便秘型肠易激综合征(IBS-C)模型小鼠5-羟色胺(5-HT)信号系统的影响,为临床治疗提供依据。方法:36只野生型小鼠,按随机数字表法分为3组,每组12只。对照组不做处理,模型组与电针组经冰水灌胃法成功建立IBS-C模型。电针组针刺双侧天枢穴、太冲穴,日1次,每次15 min,共治疗14 d。对照组和模型组均正常喂养,不做其他特殊处理。观察各组小鼠首粒黑便排出时间、粪便颗粒数及含水量,检测各组小鼠血清中5-HT的表达以及远端结肠组织5-羟色胺4型受体(5-HT4R)蛋白及mRNA的表达。结果:与对照组比较,模型组小鼠首粒黑便排出时间延长,粪便颗粒数及含水量降低,血清中5-HT的表达升高,远端结肠组织5-HT4R蛋白及mRNA的表达下调;与模型组比较,电针组小鼠首粒黑便排出时间缩短,粪便颗粒数及含水量增加,小鼠血清中5-HT的表达量下调,远端结肠组织5-HT4R蛋白及mRNA的表达上调。结论:电针天枢穴和太冲穴可以通过调节5-HT信号系统治疗IBS-C。

Abstract:Objective:To investigate the effect of electro-acupuncture at Tianshu point(ST25) and Taichong point(LR3) on 5-hydroxytrptamine(5-HT) signal system in the treatment of irritable bowel syndrome of constipation type(IBS-C). Methods:A total of 36 wild-type mice were divided into three groups according to the random number table method,with12 mice in each group. There was no treatment in the blank group. The mouse models with IBS-C were established in the model group and electro-acupuncture group by ice water gavage. The electro-acupuncture group was treated with acupuncture at both Tianshu and Taichong points,once a day,15 min each time,for 14 days. Both the control group and the model group were fed normally without special treatment to observe the time of the first black stool excretion,the number of fecal particles and water content,the expression of 5-HT in the serum of each group of mice,and the expressions of 5-hydroxytrptamine type 4 receptor(5-HT4 R) protein and mRNA in the distal colonic tissue.Results:Compared with the control group,the first black stool excretion in the model group was prolonged,the number of fecal particles and water content were decreased,the expression of 5-HT in serum was increased,and the expressions of 5-HT4 R protein and mRNA in distal colonic tissue were down-regulated.Compared with the model group,the first black stool excretion time was shortened,the number of fecal particles and water content were increased,the expression of 5-HT in serum was down-regulated,and the expressions of 5-HT4 R protein and mRNA in distal colonic tissue were up-regulated in the electro-acupuncture group. Conclusion:Electro-acupuncture at Tianshu point and Taichong point can treat IBS-C through regulating 5-HT signal system.
