大黄?丸对TGF-β1干预后A549细胞增殖的影响

目的基于"特发性肺纤维化(IPF)患者存在肺络干血"假说,观察大黄?虫丸含药血清对TGF-β1干预后A549细胞增殖的影响,探讨其防治特发性肺纤维化的作用机制。方法正常培养并取对数生长期A549细胞,5 ng/mL的TGF-β1对其干预后,Masson染色观察上皮-间充质转化(EMT)过程;选择吡非尼酮作为阳性对照药,采用血清药理学和cck-8法观察不同浓度大黄?虫丸含药血清对TGF-β1干预后A549细胞增殖的影响。结果成功获取发生EMT的A549细胞;24 h,OD值从高到低依次为:空白对照组(K组)、大黄?虫丸含药血清中剂量组(Z组)、大黄?虫丸含药血清小剂量组(X组)、模型组(M组)、大黄?虫丸含药血清大剂量组(D组)、西药组(P组);48 h,OD值从高到低依次为:K组、M组、Z组、X组、P组、D组;72 h,OD值从高到低依次为:K组、M组、D组、Z组、P组、X组。结论大黄?虫丸含药血清对TGF-β1干预后A549细胞增殖率有影响,推测大黄?虫丸对IPF的保护机制可能与不同程度阻断或逆转了TGF-β1诱导的EMT有关。

Effect of Dahuang Zhechong Pills on A549 cell proliferation after TGF-β_1 intervention

Objective Based on the hypothesis of "the presence of pulmonary collateral hard blood in IPF patients",observe the effect of medicated serum in Dahuang Zhechong Pills on the proliferation of A549 cells after TGF-β1 intervention,and explore its mechanism in preventing and treating idiopathic pulmonary fibrosis (IPF).Methods A549 cells were cultured under normal conditions and chosen in the logarithmic growth phase.After 5 ng/mL TGF-β1 intervention,Masson staining was used to observe the process of epithelial-mesenchymal transition (EMT).With pirfenidone as the positive control drug,serum pharmacology and cck-8 method were used to observe the effect of medicated serum of different concentrations in Dahuang Zhechong Pills on A549 cells proliferation after TGF-β1 intervention.Results A549 cells with EMT were successfully obtained.After 24 hours,the OD values from high to low were as follows:blank control group (K group),Dahuang Zhechong Pills medicated serum with medium dose group (Z group),Dahuang Zhechong Pills medicated serum with low dose group (X group),model group (M group),Dahuang Zhechong Pills medicated serum with high dose group (D group),western medicine group (P group).After 48 hours,the OD values from high to low were as follows:K group,M group,Z group,X group,P group,D group.After 72 hours,the OD values from high to low were:K group,M group,D group,Z group,P group,X group.Conclusion Medicated serum in Dahuang Zhechong Pills has an effect on the proliferation rate of A549 cells after TGF-β1 intervention.It is speculated that the protective mechanism of Dahuang Zhechong Pills on IPF may be related to blocking or reversing the EMT induced by TGF-β1 to certain degrees.
