枳实生制品提取液中黄酮类成分及其单体在Caco-2细胞模型中的吸收转运研究

摘    要:目的比较研究枳实和麸炒枳实中黄酮类成分橙皮苷、新橙皮苷及其单体在Caco-2细胞模型中的吸收转运机制,从而解析炮制原理。方法利用Caco-2细胞模型研究枳实和麸炒枳实中黄酮类成分橙皮苷、新橙皮苷及其单体的双向跨膜转运特征,通过高效液相色谱法测定和分析枳实炮制前后橙皮苷、新橙皮苷经细胞转运后的含量变化,计算表观渗透系数(Papp),考察时间对二者吸收和转运的影响。结果麸炒枳实的橙皮苷和新橙皮苷双侧表观渗透系数值总体上大于枳实,各药液双侧转运速率均在60 min较高,之后随着时间的延长,呈现降低趋势。结论枳实和麸炒枳实中的黄酮类成分橙皮苷、新橙皮苷及其单体具有良好的肠吸收特性,麸炒能够增加枳实内这两种黄酮类成分口服后的肠吸收,且在麸炒后橙皮苷的变化较为明显。

Abstract:Objective To compare the absorption and transportation mechanisms of flavonoids(hesperidin and neohesperidin)in raw and processed products of Zhishi(Aurantii Fructus Immaturus)and their monomers in Caco-2 cells model,and analyze the processing principle.Methods The Caco-2 cells model was used to study the bidirectional transmembrane transportation characteristics of hesperidin and neohesperidin in raw and processed products and their monomers.The change of the contents of hesperidin and neohesperidin in raw and processed products after cell transportation were analyzed by high performance liquid chromatography.We calculated the apparent permeability coefficient(Papp),and studied the effect of time on the absorption and transportation of the two.Results The double-sided apparent permeability coefficient of processed products hesperidin and neohesperidin was generally higher than that of raw of Zhishi(Aurantii Fructus Immaturus),and the bilateral transportation rate of each liquid was higher at 60 min,and then decreased with time passing by.Conclusion The flavonoids(hesperidin and neohesperidin)and their monomers in raw and processed products have good intestinal absorption characteristics,and after processing,can increase the intestinal absorption of these two flavonoids after oral administration,and the change of hesperidin is more obvious after processing.
