MiR-182对间皮瘤NCI-H2452细胞增殖、凋亡、侵袭的的影响

摘    要:目的探究MiR-182对恶性胸膜间皮瘤(malignant pleural mesothelioma, MPM)细胞生物学行为的影响。方法构建miR-182的类似物miR-182 mimic。将人间皮瘤细胞NCI-H2452分为三组:Control组、NC mimic组和miR-182 mimic组,并进行相应干预。然后,RT-PCR测定各组细胞miR-182的表达,MTT法评估细胞活力,流式检测细胞增殖、凋亡情况,而Transwell检测细胞侵袭。结果 miR-182 mimic组中miR-182的表达显著高于对照组。miR-182 mimic组的细胞活力和S+G2/M期占比较对照组均显著增加,而三组细胞的凋亡水平无差异。此外,miR-182 mimic组穿过Transwell小室的细胞数亦明显增多。结论上调miR-182可促进MPM细胞的增殖和侵袭,故miR-182可能是MPM新的治疗靶点。

Abstract:Objective To investigate the influence of miR-182 on the biological behavior of malignant pleural mesothelioma(MPM)cells.Methods MiR-182 mimic(analogue of human miR-182)was constructed.Human mesothelioma cells NCI-H2452 were divided into three groups:the control group,the NC-mimic group and the miR-182 mimic group,and they were executed their own interventions.The expression of miR-182 in each group was then determined by RT-PCR,cells viability was assessed by MTT,proliferation and apoptosis were detected by flow cytometry,and invasion ability was detected by Transwell assay.Results The expression of miR-182 in the miR-182 mimic group was significantly higher than that in the control group.The cell viability and the sum of S and G2/M phases in the miR-182 mimic group were significantly higher than those in the control group.There was no significant difference in cell apoptosis among the three groups.In addition,the number of cells passing through the transwell membrane in the miR-182mimic group was more than the other two groups.Conclusion Up-regulation of miR-182 can enhance the proliferation and invasion ability of MPM cells,therefore miR-182 may be a novel therapeutic target of MPM.
