响应面法优化丹参有效成分的提取工艺及其体外抗氧化活性研究

摘    要:目的应用响应面法优化丹参中丹参酮ⅡA和丹酚酸B两类有效成分的集成提取工艺,并对其体外抗氧化活性进行研究。方法应用高效液相色谱法同时检测丹参酮ⅡA和丹酚酸B的含量,并以此为评价指标,应用响应面法优化丹参中丹参酮ⅡA和丹酚酸B的集成提取工艺条件。此外,测定提取样品对DPPH和ABTS自由基的清除活性。结果响应面法得到的最优提取工艺条件为:12倍量67%乙醇,提取2次,每次48 min。体外抗氧化实验表明,应用此工艺提取得到的丹参提取物可剂量依赖性地清除DPPH和ABTS自由基,其清除能力优于Vc。结论响应面法用于丹参中丹参酮ⅡA和丹酚酸B的集成提取工艺优化,预测性良好,此工艺得到的丹参提取物具有较好的抗氧化活性,在氧化应激相关疾病的防治中值得进一步的研究开发。

Abstract:Objective To optimize the integrated extraction technology for extracting tanshinoneⅡA and salvianolic acid B of Danshen(Salvia miltiorrhiza).Methods The contents of tanshinoneⅡA and salvianolic acid B were determined by high performance liquid chromatography(HPLC),and the integrated extraction technology was optimized by response surface methodology.Meanwhile,the scavenging effects of integrated extracts on DPPH and ABTS were determined.Results TanshinoneⅡA and salvianolic acid B could be efficiently extracted by response surface method at the same time.The optimal extraction conditions were as follows:the ethanol concentration was 67%,the solid-liquid ratio was 1∶12,and the extraction time was 48 min.Conclusion The simultaneous extraction of tanshinoneⅡA and salvianolic acid B by response surface methodology can save time,labor and energy,and optimize the extraction technology of Danshen(Salvia miltiorrhiza).It is reasonable,feasible and suitable for the modern production.It may be useful for further research and prevention and therapy of oxidative stress-related diseases.
