不同频率水沟穴提插对局灶性脑缺血再灌注大鼠的影响

摘    要:目的探讨不同频率提插水沟穴对局灶性脑缺血再灌注大鼠的影响。方法选取健康成年Wistar大鼠78只,随机分为正常组、假手术组、模型组、低频组、中频组和高频组,每组13只,其中模型组、低频组、中频组和高频组建立局灶性脑缺血再灌注大鼠,低频组、中频组和高频组分别给予针刺干预,观察各组大鼠神经行为学,检测脑细胞凋亡指数、血清肿瘤坏死因子-α(Tumor Necrosis Factor,TNF-α)、白细胞介素-1β(Interleukin-1β,IL-1β)和白细胞介素-10(interleukin-10, IL-10)以及大脑皮质Caspase-3、PCNA表达。结果模型组、低频组、中频组和高频组大鼠行为学评分、脑细胞凋亡指数、血清TNF-α、IL-1β和IL-10、大脑皮质Caspase-3、PCNA阳性细胞比例明显高于正常组和假手术组(P<0.05),其中高频组大鼠行为学评分、脑细胞凋亡指数、血清TNF-α、IL-1β和IL-10、大脑皮质Caspase-3、PCNA阳性细胞比例分别为(4.01±0.24)分、(8.56±1.14)%、(150.06±30.15)pg·mL-1、(16.60±2.11)pg·mL-1、(37.74±3.20)pg·mL-1、(39.64±9.70)%和(12.26±1.90)%,明显低于低频组、中频组和模型组(P<0.05)。结论不同频率提插水沟穴有助于局灶性脑缺血再灌注大鼠恢复,其中针刺高频组效果较好,可能与降低大脑皮质细胞Caspase-3、PCNA表达有关。

Abstract:Objective To investigate the effects of different frequency of lifting and inserting Shuigou(DU26) on focal cerebral ischemia reperfusion in rats. Methods Seventy-eight healthy adult Wistar rats were randomly divided into normal group, sham-operated group, model group, low-frequency group, medium-frequency group and high-frequency group with 13 rats in each group. The focal cerebral ischemia-reperfusion rats were established in model group, low-frequency group, medium-frequency group and high-frequency group. The low-frequency group, middle-frequency group and high-frequency group were given acupuncture intervention respectively. The neurobehaviology of rats in each group was observed, the apoptotic index of brain cells, expressions of serum TNF-α, IL-1β and IL-10, Caspase-3 and PCNA in cerebral cortex were detected. Results The behavioral score, apoptotic index, expressions of serum TNF-α, IL-1β and IL-10, Caspase-3 and PCNA positive cells in model group, low-frequency group, medium-frequency group and high-frequency group were significantly higher than those in the normal group and sham-operated group(P<0.05). The behavioral score, apoptotic index of brain cells, expressions of serum TNF-α, IL-1β and IL-10, the percentage of Caspase-3 and PCNA positive cells in the high frequency group were(4.01±0.24)points,(8.56±1.14)%,(150.06±30.15)pg·mL-1,(16.60±2.11)pg·mL-1,(37.74±3.20)pg·mL-1,(39.64±9.70)% and(12.26±1.90)%, which were significantly lower than those of the low frequency group, the medium-frequency group and the model group(P<0.05). Conclusion Different frequencies of lifting and inserting Shuigou(DU26) are helpful to the recovery of rats with focal cerebral ischemia reperfusion. Acupuncture at high frequencies has better effect, which may be related to the decrease of expressions of Caspase-3 and PCNAin cerebral cortex cells.
