运动调节小凹蛋白-1表达改善2型糖尿病小鼠骨髓内皮祖细胞迁移能力

摘    要:目的:探究不同运动方式是否调节小凹蛋白-1(caveolin-1,Cav-1)表达及其对2型糖尿病小鼠骨髓内皮祖细胞(endothelial progenitor cells,EPCs)迁移能力的影响。方法:将40只8周龄雄性db/db小鼠按电脑产生的随机数字进行编号,随机分为有氧运动组、抗阻运动组、联合有氧+抗阻运动组(联合运动组)以及对照组,每组10只。经过8周的运动干预后分离小鼠骨髓细胞,运用密度梯度离心法分离骨髓EPCs,并以EGM-2MV培养基进行体外培养,采用双荧光染色阳性率和流式细胞仪检测表型CD133共鉴定EPCs。采用细胞划痕实验反映细胞迁移情况,Western blot检测各组Cav-1蛋白表达水平。结果:与对照组相比,有氧运动、抗阻运动及联合运动组迁移能力都明显增强(P<0.05),且联合运动组效果最佳(P<0.05)。联合运动组的Cav-1表达水平高于其它三组(P<0.05)。结论:联合运动较单纯有氧、抗阻运动及对照组更能上调Cav-1的表达,且明显改善2型糖尿病小鼠EPCs的迁移能力。

Abstract:Objective To explore whether aerobic exercise,resistance exercise,or their combination regulates the expression of small concave protein 1 c(caveolin-1,Cav-1) and the migration ability of endothelial progenitor cells(EPCs) of the bone marrow in the type 2 diabetes mice. Methods Forty 8-week-old male db/db mice were randomly divided into an aerobic exercise group,a resistance exercise group,a combined exercise group and a control group,each of 10.After 8-week exercise,bone marrow cells were isolated,from which EPCs were isolated using the density gradient centrifugation and cultured in vitro with EGM-2 MV medium.The positive rate of double fluorescence staining and phenotype CD133 detected by flow cytometry were used to identify EPCs. The cell migration was assessed using the scratch test,while the expression level of Cav-1 protein was detected using the Western blotting. Results Compared with the control group,the migration ability of EPCs of the other 3 groups was significantly enhanced(P<0.05),and the combined exercise group had the best effect(P<0.05).The expression level of Cav-1 of the combined exercise group was significantly higher than the other 3 groups(P<0.05).Conclusion The combined exercise is superior to single aerobic exercise or resistance exercise in improving the expression level of Cav-1 and the migration ability of EPCs of type 2 diabetic mice.
