索拉非尼对肾癌786-O细胞株体外培养的增殖抑制作用

目的探讨索拉非尼对体外培养的肾癌786-O细胞株细胞增殖、细胞周期和凋亡的影响。方法体外培养人肾癌786-O细胞株，给予不同浓度（10．0、50、25、1．0、0．5μmol／L）的索拉非尼混合处理，采用MTT法检测细胞增殖的变化，并采用流式细胞仪检测索拉非尼对786-O细胞凋亡和细胞周期变化的作用。结果经索拉非尼处理后，镜下可见786—O细胞株细胞结构消失，各孔内可见大量紫黑色针状结晶物，但随着药物浓度的升高，针状结晶物逐渐减少。与对照组比较，10．0、5．0和2．5μmol／L组吸光度值均显著下降（均P〈0．01）、细胞抑制率均显著升高（均P〈001），100和5．0umol／L组细胞存活率均显著下降（均P〈0．01）。与对照组比较，5．0、2．5、1．0μmol／L组G0／G1期细胞所占比例均显著升高（均P〈0．01）、S期细胞所占比例均显著下降（均P〈0．01）；而GJM期细胞所占比例仅5．0μmol／L组显著升高（P〈0．01）。结论索拉非尼可以抑制人肾癌786-O细胞株的增殖，诱导肾癌细胞凋亡。

Inhibitory effect of sorafinib on proliferation of human renal carcinoma 786-O cell line in vitro

Objective To investigate the proliferation-inhibitory effect of sorafinib on human renal carcinoma 786-0 cell line in vitro. Methods The cultured human renal carcinoma 786-0 cells were treated with different doses of sorafinib; the cell proliferation was measured by MTT assay and inhibition ratio was calculated. Cell apoptosis and cell cycle were evaluated by flow cytometry. Results After treatment of sorafinib （10, 5, 2.5, 1, 0.5μmol/L） the cellular structure disappeared and the purple-black needle-shaped crystals were observed under light microscope; the amount of crystal gradually reduced with the elevation of sorafenib concentration. MTT assay demonstrated that cell inhibition rates increased with the elevation of drug concentration （P〈0.01）. Compared with control, the ratio of G0/G1 cell of 5.0, 2.5 and 1.0μmol/L group increased significantly （P 〈 0.01）, the ratio of S stage cell decreased （P 〈 0.01）; while the ratio of G2/M stage cell only increased in the 5.0μmol/L sorafenib group. Conclusion Sorafinib can inhibit the proliferation of human renal carcinoma 786-0 cell line and induce its apoptosis.
