黄连素诱导人乳腺癌MCF-7细胞凋亡及其相关的氧化应激机制

摘    要:目的已有较多研究表明黄连素对许多肿瘤有治疗作用。文中探讨黄连素诱导人乳腺癌MCF-7细胞凋亡及其相关的氧化应激机制。方法 MTT法检测不同浓度黄连素(10、25、50、75、100μmol/L)对细胞活力的影响,Hochest33342检测不同浓度黄连素(10、25、50、75、100μmol/L)对细胞凋亡的影响;2,7-二氯荧光黄双乙酸盐(2’,7’-dichlorofluorescein diace-tate,DCFH-DA)荧光探针检测黄连素(50μmol/L)处理后MCF-7细胞内活性氧簇(reactive oxygen species,ROS)的变化;5,5’,6,6’-四氯-1,1’,3,3’-四乙基苯丙咪唑羰花青碘化物(JC-1)荧光探针检测黄连素处理后MCF-7细胞线粒体膜电位(mitochon-drial membrane potential,△Ψm)的变化。结果黄连素作用MCF-7细胞48 h,呈浓度依赖性致MCF-7细胞活力降低,诱导其凋亡。黄连素作用诱导MCF-7细胞内ROS大量生成并伴随△Ψm降低。结论黄连素具有抑制乳腺癌细胞活力,诱导其凋亡的作用,黄连素可能通过对乳腺癌细胞内氧化还原状态的调节,诱导ROS大量生成,从而损伤线粒体功能,诱导△Ψm降低,参与抗肿瘤作用。

Abstract:Objective Accumulative evidence shows that berberine plays an anti-cancer role in various cancer cells.This study aims to investigate berberine-induced apoptosis of breast cancer MCF-7 cells,and its related oxidative stress mechanism.Methods Human breast cancer MCF-7 cells were treated with berberine at the concentrations of 10,25,50,75 and 100 μmol/L.At 4h after the treatment,the motility of the MCF-7 cells was examined by MTT assay,their apoptosis detected by Hochest33342 staining,the levels of reactive oxygen species(ROS) in them determined by DCFH-DA staining,and the mitochondrial membrane potential(△Ψm) detected by molecular probe JC-1.Results Berberine inhibited the motility and induced apoptosis of the MCF-7 cells in a concentration-dependent manner.It also significantly elevated the ROS level and reduced △Ψm in the MCF-7 cells.Conclusion Bererine can inhibit the motility and induce the apoptosis of MCF-7 cells through regulating oxidation reduction.
